Antioxidative activity and immunological study of three kinds of mantidis ootheca before and after processing
Objective:To compare the effects of the raw and salt-processed products of three kinds of mantidis ootheca on immune function of mice and in vitro antioxidant activity.Methods:1,1-diphenyl-2-picrylhydrazyl and 2,2'-azinobis-3-ethylbenzthiazoline-6-sul phonate methods were used to investigate the changes of antioxidant activity in vitro before and after salt-processing of three kinds of mantidis ootheca.The immunocompromised mouse model was established by intraperitoneal injection of cyclophosphamide,and the weight change rate,immune organ index and kidney index were determined.Serum levels of interleukin-2(IL-2),tumor necrosis factor-α(TNF-α),interferon-γ(IFN-γ),and immunoglobulin M(lgM)were detected by enzyme-linked immunosorbent assay.Hematoxylin-eosin(HE)staining was used to observe the pathological changes of the thymus,spleen and kidney of the mice in each group.The relative expressions of nuclear transcription factor-κB p65[nuclear factor kappa-B p65,NF-κB(P65)],NF-κB(PP65),and IKB-α in mouse spleen tissues were detected by Western blot.Results:The in vitro antioxidant assay showed that the three kinds of mantidis ootheca and their salt-processed products had certain free radical scavenging effects.The anti-oxidative activity of salt-processed Tenodera sinensis Saussure was the strongest in vitro,and its IC50 values for scavenging DPPH and ABTS free radicals were 0.507 and 0.705 mg·mL-1,respectively.The results of immune activity study showed that the three kinds of mantidis ootheca and salt-processed products could improve the body weight change rate,thymus index and spleen index of the immunocompromised model mice to varying degrees.Salt-processed Tenodera sinensis Saussure had the best effect.The body weight change rate and spleen index were significantly different(P<0.05,P<0.001,P<0.01).The contents of cytokines IL-2,TNF-α,IFN-γ and immunoglobulin IgM in the model group were decreased to different degrees(P<0.001).Compared with the model group,the contents of IL-2 in the raw and salt-processed products of Tenodera sinensis Saussure were significantly adjusted back(P<0.05),and the contents of TNF-α in the three kinds of raw and salt-processed products of mantidis ootheca significantly increased(P<0.05,P<0.01,P<0.001),the contents of IFN-γ in salt-processed Tenodera sinensis Saussure,salt-processed Statilia maculata(Thunberg)and Tenodera sinensis Saussure increased,with significant differences(P<0.05,P<0.01),the content of IgM in Tenodera sinensis Saussure and salt-processed Tenodera sinensis Saussure significantly increased(P<0.001).The thymus,spleen and kidney in the model group showed different degrees of histopathological changes.Salt-processed Tenodera sinensis Saussure could improve the damage of thymus,spleen and kidney,and the effect was the best among the three kinds of mantidis ootheca and salt-processing products.The relative expressions of NF-κB(P65),NF-κB(PP65),and IKB-α were regulated back to normal by mantidis ootheca(P<0.001,P<0.000 1).Conclusion:The raw and salt-processed products of three kinds of mantidis ootheca can improve the immune function of mice,and have antioxidant activity in vitro,and the salt-processed products are better than the raw products.Salt-processed Tenodera sinensis Saussure has the strongest ability to scavenge free radicals and the best recovery effect on the immunocompromised mice caused by cyclophosphamide.
