Research progress on the effect of glycosylation on vaccine immunogenicity
Glycosylation modification is an important post-translational modification of proteins. Most proteins on cell membrane are glycoproteins, and different types of glycosylation modification reactions constantly occur inside the cell.Glycoproteins are involved in many life processes of the host, including cell growth, mutual recognition between cells, immune response, virus invasion and inflammatory response. With the technology improvement of proteomics and glycoomics, studies on glycosylation and modification of proteins have been paid more and more attention. With continuous development of biotechnology and research methods,we are having clearer understanding of the changes in protein function and configuration caused by glycosylation modification. The problems of glycotype analysis and the proportion of different glycotypes on glycoproteins are being solved with continuously developed experimental techniques such as the detection, separation and purification of glycoproteins, the determination of glycosylated amino acid sites, and the analysis of glycochain structure. There have been many studies on the influence of glycosylation on the immunogenicity of multiple vaccine/candidate antigens.Vaccine antigens/candidate antigens such as hepatitis B virus, human immunodeficiency virus(HIV),influenza virus and novel coronavirus(COVID-19)are all modified with varying degrees of glycoylation.Glycosylation modifications are also important in influencing these vaccine antigens. This article reviews the research methods of glycosylation and the effects of glycosylation on the immunogenicity of various vaccines.
