Progress in pharmacological studies on glycyrrhizic acid to attenuate inflammation-mediated diseases by inhibiting HMGB1
High-mobility group protein box1(HMGB1)is a highly conserved nuclear nonhistone chromosome interacting protein.Under physiological conditions,HMGB1 plays important roles in the modulation of DNA transcription,replication,repair,recombination,and nucleosome formation.In pathological state,excessive quantities of extracellular HMGB1 exerts cytokine-like effects and mediates inflammatory response.HMGB1 has been reported to be involved in a variety of inflammatory pathologies including sepsis,ischemia reperfusion injury,traumatic injury,immune disorders and so on.Glycyrrhizic acid(GL)is a triterpene saponin isolated from the roots and rhizomes of Glycyrrhiza spp.Increasing studies have demonstrated that GL exhibited multifaceted pharmacological functions.Recent studies showed that GL displayed anti-inflammatory activities via binding directly to HMGB1 and inhibiting the interaction of extracellular HMGB1 with its receptors.Here we summarize the therapeutic potential of GL and its derivatives in HMGB1-mediated inflammatory diseases,which would provide considerable experimental foundation for future clinical application of GL.
