摘要

目的:通过网络药理学结合多指标正交实验优选颈眩宁方的提取工艺.方法:采用网络药理学结合分子对接筛选颈眩宁方的潜在活性成分及靶点,并参考《中华人民共和国药典》2020年版确定指标成分.运用信息熵权法赋权及正交实验设计,以加水倍量、提取时间、提取次数为影响因素,以指标成分转移率为评价指标,确定颈眩宁方最佳提取工艺.结果:确定指标成分为天麻素、对羟基苯甲醇及葛根素,最佳提取方法为提取2次,每次提取时间1.5h,第1次加水12倍,第2次加水10倍.结论:通过网络药理学及分子对接技术,确定工艺评价指标,优选的颈眩宁方提取工艺合理、稳定、重复性良好,可为后续深入研究提供参考.

Abstract

Objective:To optimize the extraction process of Jing Xuan Ning by network pharmacology combined with multiple index orthogonal test.Methods:Network pharmacology combined with molecular docking was used to screen the potential active ingredients and targets of Jing Xuan Ning.The index components were identified according to the Chinese Pharmacopoeia(2020 edition).Using the information entropy weighting method and orthogonal test design,the optimal extraction process of Jing Xuan Ning was evaluated using transfer rate of the index ingredient,taking water addition times,extraction duration and extraction times as the influencing factors.Results:The index components were identified as gastrodin,4-hydroxybenzyl alcohol and puerarin.The optimum extraction method was to extract twice for 1.5 h each,adding 12 times of water the first time and 10 times of water the second time.Conclusion:By using network pharmacology and molecular docking techniques,the process evaluation indexes were determined,and the optimized extraction process of Jing Xuan Ning was reasonable,stable and well reproducible,which can provide a reference for subsequent in-depth research.
