基于特征图谱和量值传递评价经典名方保元汤颗粒剂的关键生产工艺

目的:基于特征图谱和 4 种指标成分的转移率评价经典名方保元汤(Baoyuan Decoction,BYD)颗粒剂的关键生产工艺,确保颗粒剂与基准样品的等效性.方法:建立BYD颗粒剂中人参皂苷Rb1、毛蕊异黄酮-7-O-葡萄糖苷、甘草酸、甘草苷的含量测定方法.以BYD基准样品为参比,建立3 批中试样品的特征图谱,评价BYD颗粒剂制备工艺的合理性.结果:建立的4 种指标成分含量测定方法稳定可行.与基准样品相比,人参皂苷Rb1、毛蕊异黄酮-7-O-葡萄糖苷、甘草酸、甘草苷从饮片到颗粒剂平均转移率分别为76.02%,63.50%,66.99%,83.54%,4 种成分的转移率均在基准样品的70%~130%范围内,且均存在对应特征峰.结论:通过对特征图谱和指标成分在各环节量值传递的研究,为今后经典名方BYD颗粒剂生产和质量控制提供参考依据.
Evaluation of key production process of Baoyuan Decoction Granules based on characteristic chromatograms and quantitative transfer relationship
Objective:To evaluatehe the key production processes of the classic famous prescription Baoyuan Decoction(BYD)Granules based on the characteristic chromatogram and the transfer rates of 4 index components to ensure the equivalence of the granules with the reference sample.Methods:A method was established for the determination of ginsenoside Rb1,calycosin-7-O-glucoside,glycyrrhizic acid and liquiritin in BYD Granules.The characteristic chromatograms of three batches of pilot samples were established with BYD reference samples as reference,and the rationality of the preparation process of BYD Granules was evaluated.Results:The established method for content determination of the four index components was stable and feasible.Compared with the reference samples,the average transfer rates of ginsenoside Rb1,galycosin-7-O-glucoside,glycyrrhizic acid and liquiritin from the tablets to the granules were 76.02%,63.50%,66.99%and 83.54%,respectively,and the transfer rates of the four components were within the range of 70%~130%of the reference samples,and all of them had corresponding characteristic peaks.Conclusion:Through the study of the characteristic chromatogram and the transfer of the index components in each link,it provides a reference basis for the future production and quality control of the classic famous prescription BYD Granules.
