Exploration of the lactogenic effect of Hordei Fructus Germinatus water decoction on hypogalactia model rats based on the interaction between intestinal flora and related hormone factors
Objective:To investigate the effects of Hordei Fructus Germinatus(HFG)water decoction on body weight,prolactin level,hormone level,mammary gland tissue,and intestinal microflora in agalactia model rats.Methods:The agalactia model was established by adjusting the number of rats in each litter.SD rats were randomly divided into five groups according to their body weight:model group,control group,and low,medium,and high-dose HFG decoction group,with 10 rats in each group.The model group was gavaged with normal saline,the control group was gavaged with metoclopramide(1.870mg·kg-1),and the HFG group was given low,medium and high doses(0.900,1.800,3.600mg·kg-1)for15d.The hourly milk yield,body weight,pups weight gain of rats were determined in each group.The serum levels of prolactin(PRL),estradiol(E2),growth hormone(GH),breast histopathological,and intestinal microflora changes were measured.The relationship between the intestinal flora in vivo and the pertinent hormone variables in rat serum was examined using Pearson correlation analysis.Results:After 15 days of intervention,the medium-dose group significantly increased the body weights of mother rats and pups rats,milk yield per hour,and the serum levels of PRL,E2,and GH(P<0.05).Pathological examination showed that compared with the model group,the number of acini in the medium-dose group was significantly increased,the glandular lumen was highly filled,the interlobular fat and fibrous connective tissue were significantly reduced,and a large quantity of pink secretion was seen in each glandular lumen.According to the examination of intestinal flora,there were more Firmicutes and Bacteroidetes in the HFG water decoction than in the model group,although there were fewer Verrucomicrobia and Proteobacteria.Bacteroides_pectinophilus_group was found to be negatively correlated with PRL and E2(P<0.05)and significantly negatively correlated with GH(P<0.01);the genus Dorea was found to be positively correlated with E2(P<0.05);and the genus Vibrio was found to be negatively correlated with E2(P<0.05).HFG water decoction was mostly connected to the steroid degradation route,dopam-inergic synaptic signal transduction,and other pathways,according to Tax4Fun's function prediction.Conclusion:In hypogalactia model rats,HFG has been found to have a positive effect on lactation levels by regulating the intestinal flora structure and improving the secretion level of related hormone factors.This suggests that hormones and flora composition play a crucial role in improving lactation levels.
