目的:研究GW0742 激动过氧化物酶体增殖物激活受体β/δ(peroxisome proliferator activated receptor β/δ,PPARβ/δ)对新生大鼠生发基质出血(germinal matrix hemorrhage,GMH)的神经保护作用,并探讨其作用机制.方法:7 日龄乳鼠按体质量随机分为假手术组(Sham,S),模型组(GMH,G),GW0742 低剂量组(low-dose,L)、中剂量组(medium-dose,M)、高剂量组(high-dose,H).右半球生发基质层注射0.3U胶原酶诱导GMH模型,造模后30 min腹腔注射GW0742.神经行为学测试评估SD乳鼠的神经功能;脑含水量评估其脑水肿程度;脑血红蛋白含量检测脑出血情况;试剂盒检测亚铁离子含量;HE染色观察SD乳鼠脑组织病理形态;Western blot实验检测转铁蛋白受体1(transferrin receptor 1,TFR1)、谷胱甘肽过氧化物酶4(gluta-thione peroxidase 4,GPX4)蛋白表达.结果:与S组比较,G组翻正反射实验和负趋地性实验消耗时间增加,神经功能障碍评分升高(P<0.01,P<0.01,P<0.01),脑含水量、脑血红蛋白含量、脑组织亚铁含量增加(P<0.01,P<0.01,P<0.01),患侧脑组织损伤显著,上调TFR1 蛋白表达(P<0.05),下调GPX4 蛋白表达(P<0.05);与G组比较,L组缩短翻正反射实验和负趋地性实验消耗时间,评分降低(P<0.05,P<0.05,P<0.05);脑含水量和脑血红蛋白含量以及脑亚铁含量降低(P<0.05,P<0.05,P<0.05).M组翻正反射实验和负趋地性实验消耗时间减少(P<0.01,P<0.01),评分降低(P<0.01);脑含水量和脑血红蛋白含量以及脑亚铁含量降低(P<0.01,P<0.01,P<0.01).H组翻正反射实验和负趋地性实验消耗时间减少(P<0.01,P<0.01),评分降低(P<0.01),脑含水量和脑血红蛋白含量以及脑亚铁含量降低(P<0.01,P<0.01,P<0.01),脑组织损伤减轻,TFR1 蛋白表达减少(P<0.05),GPX4 蛋白表达增加(P<0.01).结论:GW0742 激动PPARβ/δ对乳鼠生发基质出血具有神经保护作用.
Agitation of peroxisome proliferator activated receptor β/δ by GW0742 protected against germinal matrix hemorrhage in neonatal rats
Objective:To investigate the neuroprotective effect of GW0742 through activating peroxisome proliferator activated receptor β/δ against germinal matrix hemorrhage in neonatal rats and to explore its mechanism of action.Methods:Seven-day-old rat pups were randomly assigned to Sham,GMH,low-,medium-,and high-dose groups according to their body weight.GMH was induced by injection from the dura mater of collagenase(0.3 U)into the right hemispheric germinal matrix,and GW0742 was peritoneally injected 30 min postoperatively.Neurobe-havioral examinations were employed to evaluate the nervous function of SD neonatal rats;brain water content to assess the degree of cerebral oedema;hemoglobin assay to detect cerebral hemorrhage;ferrous ion detection kit to detect the ion contents;HE staining to observe the pathological morphology of brain tissues of SD neonatal rats;and Western blot experiment to detect the expression of transferrin receptor 1(TFR1)and glutathione peroxidase 4(GPX4)protein expression.Results:Compared with group S,group G showed increased time consumption and higher neurological dysfunction scores in the body righting test and the negative geotaxis test(P<0.01,P<0.01,P<0.01),and increased brain water contents,brain blood volume,and brain tissue ferrous iron contents(P<0.01,P<0.01,P<0.01),and significantly destroyed brain tissues,upregulated TFR1 protein expression(P<0.05)and downregulated GPX4 protein expression(P<0.05)were also found.Compared with group G,group L showed decreased time consumption and lower neurological dysfunction scores in the body righting test and the negative geotaxis test(P<0.05,P<0.05,P<0.05),and decreased brain water contents,brain blood volume,and brain tissue ferrous iron contents(P<0.05,P<0.05,P<0.05);group M showed decreased time consumption and lower neurological dysfunction scores in the body righting test and the negative geotaxis test(P<0.01,P<0.01,P<0.01),and decreased brain water contents,brain blood volume,and brain tissue ferrous iron contents(P<0.01,P<0.01,P<0.01);group H showed decreased time consumption and lower neurological dysfunction scores in the body righting test and the negative geotaxis test(P<0.01,P<0.01,P<0.01),and decreased brain water contents,brain blood volume,and brain tissue ferrous iron contents(P<0.01,P<0.01,P<0.01),and reduction of brain tissue damage,downregulated TFR1 protein expression(P<0.05)and upregulated GPX4 protein expression(P<0.01)were also found.Conclusion:Activation of PPARβ/δ by GW0742 indicated neuroprotective effect against germinal matrix hemorrhage in neonatal rats.
