Relationship among polymorphisms at rs9939609 locus and rs1421085 locusof fat-mass and obesity-as-sociated gene and blood lipid level in elderly patients with hyperlipidemia
Objective To discuss the relationship among polymorphisms at rs9939609 locus and rs1421085 locusof fat-mass and obesity-associated gene(FTO)and blood lipid level in elderly patients with hyperlipidemia.Methods The elderly patients with hyperlipidemia(n=104)were chosen into case group and healthy elderly people(n=91)were chosen into control group from Department of General Practice in Affiliated Hospital of North China University of Science and Technology from Jan.2021 to Dec.2022.The general materials were collected,and samples of fasting peripheral venous blood in the morning to detect the levels of lipid and other biochemical indexes.The rs9939609 locus and rs 1421085 locus of FTO gene were typed by using Sanger sequencing.The general materials,genotypes,gene frequency and blood lipid level in patients with different genotypes were compared in 2 groups.Results The levels of body mass index(BMI),waist hip ratio(WHR),triglyceride(TG),total cholesterol(TC),low-density lipoprotein(LDL-C)and apolipoprotein B100(ApoB100)were higher in case group than those in control group(P<0.05).The frequency of genotypes TA and AA at rs9939609 locus were higher(P=0.025)andfrequency of allele A was higher(P=0.013),and genotypes TC and CC at rs1421085 locus were higher(P=0.025)and frequency of allele C was higher(P=0.013)in case group.There was difference in TG,high-density lipoprotein(HDL-C)and ApoA1 among different genotypes at rs9939609 locus(P<0.05).The level of HDL-C was lower in genotype TA than that in genotype TT(H=20.68,P=0.003).There was difference in TG,HDL-C and ApoA1 among different genotypes at rs1421085 locus(P<0.05),and HDL-C was lower in genotype TC than that in genotype TT(H=20.68,P=0.003).Conclusion The base mutationsat rs9939609 locus and rs1421085 locusof FTO may increase the risk of dyslipidemia in the elderly population,which is related to TG and HDL-Cmetabolism.
