Concentration Determination of Clozapine and Quetiapine in Human Serum by HPLC
OBJECTIVE:To establish a method for simultaneous determination of clozapine and quetiapine concentration in human serum.METHODS:After alkalizing by 0.01 mol/L NaOH,human serum samples were extracted by n-pentane and analyzed by HPLC.The determination was performed on SB-C18 column with mobile phase consisted of methanol-water-tetramethylethylenediamine-acetic acid (620∶380∶4.4∶3.52) at the flow rate of 0.8 ml/min.The column temperature was 40 ℃ and UV detection wavelength was set at 254 nm.Injection volume was 20 μl,and 2-amino-5-chloro-2'-fluoro benzophenone was used as internal standard.RESULTS:The linear range were 10-1 200 μg/L for clozapine (r=0.998 5) and 10-700 μg/L for quetiapine (r=0.996 3),respectively,the quantitative lower limit were both 10 μg/L.The extraction recoveries of clozapine and quetiapine at high,middle and low concentrations were greater than 70%.The intra-day and inter-day RSD were less than 15% (n=5).CONCLUSIONS:The method is sensitive,simple,rapid and accurate,and can be used for the determination of clozapine and quetiapine concentration in human serum.
