In order to explore the inhibitory effect of water extracts of five traditional Chinese medicines(Dandelion,Isatis root,Alisma orientalis,Kudzu root and Scutellaria baicalensis)on porcine epidemic diarrhea virus(PEDV)infection in vitro,in this study,different concentrations of traditional Chinese medicine solutions were first added to African green monkey kidney cells(Vero cell),and the impact on cell growth status was observed to determine the maximum safe concentration of drugs.The results showed that the maximum safe concentrations of Dandelion,Isatis root,Rhizoma alismatis,Kudzu root and Scutellaria baicalensis were 3.91mg/mL,1.95mg/mL,1.95mg/mL,0.98mg/mL and 0.98mg/mL,respectively.Use the maximum safe concentration of the drug as the original solution,and the drugs were diluted by two-fold serial dilution method.Three different methods were used to add the drugs to Vero cell monolayer.After 72 hours,the OD450nm value of each well was measured by CCK-8 method to evaluate the concentration of each drug with the highest inhibition rate against PEDV under the blocking,inhibiting and killing effects.The results showed that the concentrations of the five drugs with the highest inhibition rates were 3.91mg/mL,1.95mg/mL,1.95mg/mL,0.98mg/mL,and 0.98mg/mL,respectively.In order to evaluate the inhibitory rate of each drug against PEDV under the three modes of action,the drug with the highest inhibitory rate against PEDV was selected,and cell viability was measured using the CCK-8 method.The results showed that all traditional Chinese medicines could inhibit the infection of PEDV in vitro.Under the blocking effect,the inhibition rate of Scutellaria baicalensis on PEDV reached 138.10%,under the inhibiting effect,the inhibition rate of Dandelion on PEDV reached 149.90%,under the killing effect,the inhibition rate of Kudzu root on PEDV reached 102.90%,and the inhibition rate of Dandelion and Scutellaria baicalensis on PEDV under the three mode of action was more than 80%.The above results showed that Dandelion and Scutellaria baicalensis have the best effect of anti-PEDV infection in vitro.This study first validated the effect of five traditional Chinese medicines on anti-PEDV infection in vitro,which can be used as a reference for the treatment of PEDV with traditional Chinese medicine.
