To understand the prevalence and drug resistance of Clostridium perfringens isolated from sheep in Shandong Province,150 fresh fecal samples were collected from three large-scale sheep farms in Tai'an,Jining,and Heze areas from November 2022 to March 2023.A total of 43 strains of C.perfringens were isolated,with a isolate rate was 28.7%(43/150).The toxin genes,drug resistance genes,and 8 pairs of housekeeping genes of C.perfringens were amplified by PCR;Drug susceptibility testing was detected by K-B drug sensitive disk method,and the correlation between drug resistance phenotype and drug resistance gene was statistically analyzed.The results showed that all isolates carried the cpa toxin gene were type A.The isolates showed a high resistance rate to tetracycline and clindamycin,with 72.1%and 67.4%,respectively.The strains resistant to 1 to 5 drugs accounts for 2.3%to 48.8%,with a multi drug-resistant rate of 27.9%.The detection rate of tetracycline resistant gene tetA(P)was 44.2%,followed by β-lactam resistant gene blaCTX-M(27.9%).The coincidence rate of drug resistance phenotype and drug resistance genes was 41.2%-100%.The 43 strains of C.perfringens were divided into 23 ST types by multilocus sequence typing(MLST),of which ST1 and ST22 were the main prevalent types.The results of this study can provide a reference for the monitoring and control of C.perfringens of sheep in Shandong,as well as the rational use of antibiotics in farms.
