白头翁汤预防禽大肠杆菌病作用机制的研究

Investigation of the mechanism of pulsatilla decoction in preventing avian colibacillosis based on network pharmacology

为初步探究白头翁汤(BTWT)对肉鸡大肠杆菌感染的作用靶点,本研究通过HERB2.0数据库筛选BTWT作用靶点,再利用疾病数据库Gene cards得到大肠杆菌感染的相关靶点,筛选出药物与疾病的交集靶点.利用STRING在线分析平台构建BTWT抑制大肠杆菌感染的蛋白相互作用(PPI)网络图并筛选核心靶点,进行蛋白质的GO功能和KEGG信号通路分析,对得到的核心靶点进行分子对接验证.结果显示,筛选到BTWT的有效成分95个,疾病靶点462个,药物与疾病相互作用的交集靶点25个,核心靶点22个.GO分析得到49个功能条目,包括生物过程41个(如巨噬细胞活性、免疫应答和炎症反应)、细胞组分4个(如细胞外空间、胞浆)、分子功能4个(如干扰素-γ受体结合、细胞因子活性).KEGG信号通路分析得到11条信号通路,涉及C型凝集素受体信号通路、Toll样受体信号通路、细胞凋亡、NOD样受体信号通路等.分子对接结果显示,γ-氨基丁酸与超氧化物歧化酶1(SOD1)、秦皮素与白介素-10(IL-10)、槲皮素与IFN-γ(干扰素-γ)对接的自由能均<0.选取60只健康肉鸡,随机分为3组,20只/组,分别为健康对照组(C组)、感染组(M组)和BTWT组(PD组),C组和M组饲喂基础日粮,PD组在肉鸡基础日粮中加入10%BTWT中药液,连续饲喂7d,M组和PD组在第8d口服大肠杆菌O78株(1.09×108 cfu),2 d后计算BTWT对大肠杆菌感染肉鸡的免疫保护率和免疫器官指数,利用ELISA试剂盒测定各组鸡血清中细胞因子、抗氧化指标的含量,光学显微镜下观察各组鸡空肠组织病变.结果显示,与M组相比,BTWT对肉鸡抵抗大肠杆菌感染具有一定的免疫保护率,保护率为20%.PD组鸡法氏囊指数和胸腺指数有所升高,但与M组差异不显著(P>0.05),脾脏指数显著降低(P<0.05);PD组鸡血清中IgA含量显著升高(P<0.05),IL-10、TNF-α和IFN-γ的含量显著降低(P<0.05);PD组鸡血清中SOD和GSH-PX水平显著升高(P<0.05),MDA水平显著降低(P<0.05),空肠的损伤情况得到缓解.以上结果表明,BTWT对肉鸡大肠杆菌感染有较好的预防作用,通过调节感染肉鸡血清中SOD、IL-10及IFN-γ的分泌水平缓解大肠杆菌感染造成肉鸡肠道的损伤,为深入研究BTWT预防大肠杆菌感染的作用机制提供了新思路.
To explore the target of pulsatilla decoction(BTWT)in preventing Escherichia coli infections in broilers,the targets of BTWT were predicted in this study through the HERB2.0 database,and the related targets of E.coli infection were screened using the disease database Gene cards,and the intersection targets of the drug and the disease were identified.STRING online analysis platform was used to construct the protein-protein interaction(PPI)network diagram and screen the core targets of BTWT for inhibition of E.coli infection,the GO function of proteins and KEGG signaling pathway analyses were carried out,and molecular docking validation was carried out for the obtained core targets.The results showed that 95 active ingredients,462 disease targets,25 intersecting targets of drug-disease interactions,and 22 core targets were screened for BTWT.GO analysis yielded 49 functional entries,including 41 biological processes(e.g.,macrophage activity,immune response,and inflammatory response),4 cellular components(e.g.,extracellular space,cytoplasm),and 4 molecular functions(e.g.,interferon-gamma receptor binding),and KEGG pathway analysis yielded 11 signaling pathways,involving C-type lectin receptor signaling pathway,Toll-like receptor signaling pathway,apoptosis,NOD-like receptor signaling pathway,etc.The molecular docking results showed that the free energy of γ-aminobutyric acid docked with SOD1,Qinpiin docked with IL-10,and quercetin docked with IFN-γ were all<0 kcal/mol.Sixty healthy broilers were selected and randomly divided into three groups of 20 in each group,which were healthy control group(Group C),infected group(Group M)and BTWT group(Group PD).The Group C and M were fed with basal diet,and the Group PD were fed with basal diet supplemented with 10%BTWT for 7 days.Groups M and PD were administrated orally with E.coli O78 strain(1.09×108 cfu)at 8 days,and after 2 days,the protection rate and immune organ index of BTWT on E.coli-infected broilers were calculated,the contents of cytokines and antioxidant indexes in serum were determined by ELISA kit,and the morphological changes of the jejunum were observed under light microscope.The results showed that BTWT provided certain protection against E.coli infection in broilers with a protection rate of 20%compared with Group M.The fasciola index and thymus index in Group PD increased,but the difference was not significant(P>0.05),and the spleen index significantly decreased(P<0.05);the serum IgA content significantly increased(P<0.05),and the IL-10,TNF-α and IFN-γ levels significantly decreased(P<0.05).Meanwhile,the levels of SOD and GSH-PX in serum significantly increased(P<0.05),the level of MDA significantly reduced(P<0.05),and the damage of jejunum was alleviated.The above results indicated that pulsatilla decoction provided a good prevention against E.coli infection in broilers,and may alleviate the damage caused by E.coli infection by regulating the levels of SOD1,IL-10 and IFN-γ,offering a new idea for the in-depth study of the mechanism of action of pulsatilla decoction in the prevention of E.colidisease.
