Genetic characteristics of hantavirus in rodents in hemorrhagic fever epidemic area in Jinan City
Objective To analyze the genotypes and genomic characteristics of hantavirus(HV)causing hemorrhagic fever with renal syndrome(HFRS)in Jinan City.Methods The whole genome sequence of HV was obtained using the second-generation sequencing technology from the lung tissue samples of HV-positive mice in Jinan City in 2021.BioEdit and MEGA 11 were used to conduct genetic evolution and homologous alignment analysis of the genome sequences,plot phylogenetic tree,and analyze nucleotide and amino acid sequence mutations.Results The whole genome sequence of two HV strains was obtained,with a total length of the genome being 11 944 bp,both of which were of Seoul virus.The Seoul HV isolates from Jinan belonged to the S3 subtype,and had high level of amino acid homology in its S gene with the isolates from Beijing in 2005(98.3%-98.6%).Conclusions The whole genome sequence of HV isolates from Jinan was obtained using the second-generation sequencing method.Amino acid sequence analysis showed high homology to the Seoul type found in other areas in China,and had obvious mutations in nucleotide and amino acid sequences compared with the vaccine strains.Therefore,it is necessary to pay close attention to changes in its pathogenicity,virulence and transmission.
