Effects of nucleotide polymorphisms of tcpA gene on environmental adaptability of toxigenic strains of Vibrio cholerae O139 strain
Objective To investigate the effect of nucleotide mutation of tcpA gene on the environmental adaptability of toxigenic strains of Vibrio cholerae O139 strain.Methods The variation of tcpA gene in O139 strains was determined by bioinformatics method,and the tcpA gene mutant was constructed by homologous recombination method.The growth and difference of wild-type and mutant strains were studied by comparing their biofilm formation,pilus formation,and growth under different nutrient concentrations.Results Mutant 1 exhibited stronger growth capability under different pH conditions,whether it was grown alone or in a competitive experiment simulating the human gastrointestinal environment.Conclusions Nucleotide mutations of tcpA gene affect the acid-base tolerance of toxigenic strains of Vibrio cholerae O139 strain.
