The preventive effect of evodiamine on vascular hypertrophy in obese rats
Aim To study the preventive effect of evo-diamine on vascular remodeling in obese rats. Methods Obese rats were induced by high-fat diet; body weight, body length, abdominal circumference, and Lee's index were measured per week. Serum and tissue calcitonin gene-related peptide ( CGRP ) contents weremeasured by ELISA method. Vascular tissue and dorsal root ganglia ( DRG ) TRPV1 expression was determined by immunohistochemistry method. Serum total cholesterol, low density lipoprotein cholesterol, high density lipoprotein cholesterol and triglyceride levels were detected . Results Evodiamine and capsaicincould significantly decrease body weight, abdominal circumference, Lee ' s index, fat mass, serum total cholesterol and triglyceride in obese rats. Evodiamine obviously reduced vascular hypertrophy in obese rats. Compared with normal rats, the expression of TRPV1 in DRG and vascular tissue was decreased significantly, and CGRP levels in serum and abdominal aorta were significantly increased in obese rats, but decreased in DRG and thoracic aorta. Evodiamine and capsaicin could increase the expression of TRPV1 in DRG and vascular tissue. Evodiamine significantly de-creased CGRP levels both in serum and abdominal aorta, also, increased CGRP contents both in DRG and thoracic aorta. Conclusions Evodiamine has a good preventive effect on vascular remodeling in high-fat diet-induced obesity, and the mechanisms may be related to regulation of CGRP and TRPV1 expression. However , the detailed mechanisms of evodiamine on vascular remodeling in obese rats should be studied further.
