Study on the relationship between the levels of miR-342-3p and miR-423-5p and glucose lipid metabolism and insulin resistance in women with gestational diabetes

目的 探究妊娠糖尿病患者miR-342-3p和miR-423-5p水平与糖脂代谢以及胰岛素抵抗的相关性.方法 选取2020年9月至2022年9月在中国福利会国际和平妇幼保健院诊治的96例妊娠糖尿病患者作为研究组,另选取同期孕检健康孕妇96例为对照组,采用实时荧光定量PCR(qRT-PCR)法检测血清miR-342-3p和miR-423-5p表达;采用Pearson法分析血清miR-342-3p和miR-423-5p相关性以及二者与糖脂代谢及胰岛素抵抗相关指标的相关性;采用Logistic回归分析妊娠糖尿病的影响因素.结果 研究组血清miR-342-3p、miR-423-5p、空腹血糖(FBG)、空腹胰岛素(FINS)、总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、胰岛素抵抗指数(HOMA-IR)显著高于对照组(P<0.05),高密度脂蛋白胆固醇(HDL-C)和胰岛β细胞功能指数(HOMA-β)显著低于对照组(P<0.05).根据Pearson相关性分析得知,血清miR-342-3p和miR-423-5p水平呈正相关(P<0.05),血清miR-342-3p和miR-423-5p水平与 FBG、FINS、TC、TG、LDL-C、HOMA-IR 呈正相关(P<0.05),与 HDL-C 和 HOMA-β 呈负相关(P<0.05).根据 Logistic 回归分析得知 FBG、FINS、TC、TG、LDL-C、miR-342-3p、miR-423-5p 高表达,HDL-C 降低是影响妊娠糖尿病的危险因素(P<0.05).结论 妊娠糖尿病患者miR-342-3p和miR-423-5p水平异常升高,二者与糖脂代谢和胰岛素抵抗相关.
Objective To investigate the correlation between miR-342-3p and miR-423-5p levels and glycolipid me-tabolism and insulin resistance in patients with gestational diabetes.Methods A total of 96 patients with gestational diabetes who were treated in the China Welfare Association International Peace Maternal and Child Health Hospital from September 2020 to September 2022 were selected as the research group,and 96 healthy pregnant women during the same period were selected as the control group,and the expression of serum miR-342-3p and miR-423-5p was detected by real-time real-time PCR(qRT-PCR).The Pearson method was used to analyze the correlation between serum miR-342-3p and miR-423-5p and the correlation between them and the indexes related to glycolipid metabolism and insulin resistance.Logistic regression was used to analyze the influencing factors of gestational diabetes.Results The serum miR-342-3p,miR-423-5p,fasting blood glucose(FBG),fasting insulin(FINS),total cholesterol(TC),triglycerides(TG),low-density lipoprotein cholesterol(LDL-C),insulin resistance index(HOMA-IR)were significantly higher than those in the control group(P<0.05),and high-density lipoprotein cholesterol(HDL-C)and islet β cell function index(HOMA-β)were significantly lower than those in the control group(P<0.05).According to Pearson correlation analysis,serum miR-342-3p and miR-423-5p levels were positively correlated(P<0.05),serum miR-342-3p and miR-423-5p levels were positively correlated with FBG,FINS,TC,TG,LDL-C,HOMA-IR(P<0.05),and HDL-C and HOMA-β(P<0.05).According to logistic regression analysis,high expression of FBG,FINS,TC,TG,LDL-C,miR-342-3p,miR-423-5p,and decreased HDL-C were risk factors affecting gestational diabetes mellitus(P<0.05).Conclusion Patients with gestational diabetes have abnormally elevated levels of miR-342-3p and miR-423-5p,which are associated with glycolipid metabolism and insulin resistance.
