目的 探讨lncRNA CRNDE对子宫内膜癌HEC-1A细胞的增殖、凋亡及侵袭的影响.方法 利用RT-qPCR比较了正常子宫内膜细胞hESC与子宫内膜癌细胞HEC-1A CRNDE的表达差异,利用shRNA-CRNDE慢病毒转染HEC-1A细胞后,分别利用CCK-8、划痕实验、小室迁移实验、基质胶侵袭实验以及流式细胞术检测转染后细胞增殖、迁移和侵袭能力以及细胞凋亡水平.结果 HEC-1A细胞CRNDE表达水平高于hESC,且其增殖、迁移和侵袭能力均高于hESC.而慢病毒转染后的HEC-1A细胞增殖、迁移和侵袭能力显著减弱,但仍略高于hESC细胞.同时,CRNDE沉默还导致了 HEC-1A细胞凋亡率明显上升.结论 CRNDE在HEC-1A细胞中高表达并与细胞增殖、迁移和侵袭能力、凋亡水平直接相关,表明其有作为子宫内膜癌诊断及治疗靶点的潜力.
Objective To investigate the effect of lncRNA CRNDE on the proliferation,apoptosis,and invasion abilities of HEC-1A cells.Methods The expression difference of CRNDE in hESC cells and HEC-1A cells was compared with RT-qPCR.After transfecting HEC-1A cells with shRNA-CRNDE lentivirus,its cell proliferation,migration and invasion abilities,and cell apoptosis levels were detected by CCK-8,transwell migration experiment,matrigel invasion experiment,and flow cytometry.Results The CRNDE expression level,proliferation,migration and invasion abilities of HEC-1A cells were higher than those of hESCs.After CRNDE was inhibited,these abilities were significantly weakened,but still slightly higher than those of hESC cells.At the same time,CRNDE silencing also led to a significant increase in the apoptosis rate of HEC-1A cells.Conclusion CRNDE is highly expressed in HEC-1A cells and is directly related to cell proliferation,migration and invasion ability,and apoptosis level,indicating that it has the potential to be a diagnostic and therapeutic target for endometrial cancer.
