宫颈癌组织中MAPK6基因表达及其临床意义

摘要
目的 探究宫颈癌组织中丝裂原活化蛋白激酶-6(MAPK6)基因表达及其临床意义.方法 选取河北省张家口市妇幼保健院2017年4月至2020年1月经病理学检查并确诊的136例宫颈癌组织及癌旁组织(与肿瘤边缘距离≥2 cm)标本为研究对象.免疫组化法、Westernblot法、实时荧光定量PCR法检测宫颈癌患者癌组织和癌旁组织中MAPK6表达情况;Kaplan-Meier法分析MAPK6表达与宫颈癌患者预后的关系;COX回归分析法探讨影响患者预后的因素.结果 宫颈癌组织中MAPK6的阳性表达率[73.53％(100/136)]、蛋白表达量(1.36±0.31)及mRNA表达水平(1.83±0.49)均显著高于癌旁组织[38.24％(52/136)、0.41±0.11、1.03±0.28](P＜0.05);不同FIGO分期、分化程度、淋巴结转移、浸润深度宫颈癌患者癌组织中MAPK6阳性表达差异显著(P＜0.05);生存曲线结果表明,MAPK6阳性表达患者3年生存率显著低于MAPK6阴性表达;FIGO分期Ⅱ期、低分化、淋巴结转移、肌层浸润深度超1/2及MAPK6阳性表达均为影响宫颈癌患者预后的危险因素.结论 MAPK6在宫颈癌组织中呈高表达,其表达与病理特征及预后联系紧密.
Abstract
Objective To investigate the the expression of mitogen-activated protein kinase-6(MAPK6)gene expres-sion in cervical cancer and its clinical significance.Methods 136 case of cervical cancer tissues and adjacent tissues(distance≥2 cm from tumor margin)diagnosed by pathological examination in Zhangjiakou Maternal and Child Health Hospital of Hebei Province from April 2017 to January 2020 were selected as the study objects.Immunohistochemical method,Western blot method and real-time fluorescence quantitative PCR were applied to detect MAPK6 expression in cancerous and paracan-cerous tissues of patients with cervical cancer.Kaplan-Meier method was adopted to evaluate the relationship between expres-sion of MAPK6 in cervical cancer tissues and prognosis.COX regression analysis was conducted to discuss the factors af-fecting patients'prognosis.Results The positive expression rate[73.53％(100/136)],protein expression level(1.36±0.31)and mRNA(1.83±0.49)expression level of MAPK6 in cervical cancer tissue were significantly higher than that in adjacent tissues[38.24％(52/136),0.41±0.11,and 1.03±0.28](P＜0.05).The difference of MAPK6 positive expression in cancer tissues of cervical cancer patients with different FIGO stage,differentiation degree,lymph node metastasis and infiltration depth were significant(P＜0.05).The results of survival curve indicated that the 3-year survival rate of patients with MAPK6-positive expression was significantly lower than that MAPK6-negative expression.COX regression analysis showed that FIGO stageⅡ,low differentiation,lymph node metastasis,＞1/2 depth of infiltration,and MAPK6 positive expression were risk factors affecting the prognosis of patients with cervical cancer.Conclusion MAPK6 is highly expressed in the tissues of patients with cervical cancer,and its expression is strongly associated with pathological features and prognosis.
