Study on the expression of Leptin and the regulation of insulin resistance and autophagy related factors in PCOS rats
Objective To explore the regulatory effects of veriratol on serum leptin expression,endoplasmic reticu-lum stress,insulin resistance and autophagy related factors in PCOS rats.Methods 39 healthy rats were randomly divided into control group,PCOS group and treatment group.The control group was fed normally,the PCOS group and treatment group were treated with PCOS model,and the treatment group was treated with resveratrol.The pathological changes of beta cells in ovary and pancreas and the expression levels of leptin,endoplasmic reticulum stress.Insulin resistance and autophagy related factors were observed.Results In the control group,there were follicles at different stages of development,and the morphol-ogy of granulosa cells was complete,showing 8-9 layers,and oocytes were visible.In the PCOS group,cystic dilated follicles were increased,the granulosa cell layer was thin,and atretic follicles and oocytes were visible.In the treatment group,the granulosa cell layer was thickened,cystic dilated follicles were reduced,and mature follicles were increased.The number of βcells in the COS group was significantly decreased,and the number of β cells in the treatment group was higher than that in the PCOS group,which was slightly lower than that in the control group.There were statistically significant differences in fasting insulin,insulin resistance,leptin,oxidative stress indices,p62,LC3Ⅰ and LC3 Ⅱ protein relative expressions among all groups.Fasting insulin,HOMA-IR,leptin,MDA,p62 and LC3 Ⅱ levels in PCOS group were higher than those in control group and treatment group,and those in treatment group were higher than those in control group.The levels of HOMA-β,HOMA-s%,GSH,SOD and LC3Ⅰ in PCOS group were lower than those in control group and treatment group,and the treatment group was lower than control group(P<0.05).Conclusion Resveratrol can down-regulate the expression of leptin in PCOS rats,improve the insulin resistance,inhibit the autophagy,and improve the pathological changes of ovarian and pancreatic β cells.
