Effects of ADSCs on endometrial repair,collagen level and PPAR-α/FAO signal after mechanical injury in rats
Objective To investigate the effects of ADSCs on endometrial repair,collagen level and PPAR-α/FAO signal after mechanical injury in rats.Methods The mechanical injury model of endometrium was established in 23 rats,and 21 rats were successfully divided into model group,ADSCs group and estrogen group randomly,with 7 rats in each group,and 7 rats without model were normal group.ADSCs cell suspension was injected into tail vein in ADSCs group,and 17β-estradiol was injected subcutaneously in estrogen group.The other groups of rats did not intervene.The pathological tissue morphology,endometrial thickness,number of glands,levels of VEGF and FGF,type Ⅰ collagen and PPAR-α/FAO signaling pathway pro-tein were observed in each group.Results The endometrial shape of rats in normal group was normal,the endometrial shape of rats in model group was the worst,and the endometrial shape of rats in estrogen group and ADSCs group was better than that in model group.The endometrial thickness and number of glands in estrogen group and ADSCs group were better than model group,and ADSCs group was significantly increased(P<0.05).The expression levels of serum VEGF and FGF in estrogen group and ADSCs group were higher than those in model group,and ADSCs group were significantly increased(P<0.05).The expression of type Ⅰ collagen,PPAR-α and FAO among all groups was higher in estrogen group and ADSCs group than in model group,and the ADSCs group was significantly increased(P<0.05).Conclusion ADSCs can improve endometrial mor-phological tissue,increase endometrial thickness,number of glands and type Ⅰ collagen,promote endometrial secretion of VEGF and FGF factors,regulate PPAR-α/FAO signaling pathway protein expression,and repair the mechanical injury of en-dometrial.
