子宫内膜异位症患者外泌体调控巨噬细胞吞噬功能及极化的实验研究

目的 探讨子宫内膜异位症外泌体对巨噬细胞吞噬功能和极化的影响.方法 分离健康人群子宫内膜间质细胞的外泌体,命名为对照(CON)组,分离子宫内膜异位症间质细胞的外泌体命名为子宫内膜异位症(EMS)组.用纳米颗粒示踪技术检测各组外泌体大小、粒径以及电势.Western blot检测CON组和EMS组中外泌体标志物CD9和CD63的表达.将CON组和EMS组的外泌体与巨噬细胞THP-1进行共培养,分别命名为CON-exosomes组和EMS-exosomes组,空白对照组(PBS-Contro l组)加入等量的PBS.用MTT实验检测三组细胞的增殖活力.流式细胞术检测各组细胞的凋亡率和巨噬细胞M1极化(CD86+)细胞和M2极化(CD206+)的比例.用CM-Dil dye染料标记外泌体用DAPI标记细胞核,用激光共聚焦显微镜观察巨噬细胞对外泌体的吞噬能力.结果 CON组和EMS组外泌体的物理参数之间的差异无统计学意义(均P>0.05),两组直径分别为95.67~119.25 nm,94.82~121.33 nm(P>0.05).粒径分布的峰尺寸分别为103.46 nm和106.89 nm.CON组外泌体的Zeta电势为(-29.28±3.91)mV,EMS组外泌体的Zeta电势为(-28.64±4.52)mV(P>0.05).两组的外泌体标志物CD9、CD63的表达差异均无统计学意义(P>0.05).与PBS-Control组比,与CON-exosomes组的THP-1细胞增殖活力和M2极化比例减少(P<0.05),THP-1细胞凋亡率变化差异无统计学意义(P>0.05).与CON-exosomes组比,EMS-exosomes组的细胞增殖活力和M2极化比例增加(P<0.05),细胞凋亡率增加(P<0.05).与CON-exosomes组比,EMS-exosomes组的THP-1细胞对外泌体的吞噬功能减弱.结论 子宫内膜异位症来源的外泌体抑制THP-1细胞增殖活力和M2极化,促进THP-1细胞的凋亡,并减弱其吞噬功能.
Experimental study on the regulation of phagocytic function and polarization of macrophages by exosomes from patients with endometriosis
Objective To explore the effects of exosomes derived from endometriosis on phagocytic function and polarization of macrophages.Methods Exosomes were isolated from the stromal cells of healthy individuals,named the con-trol(CON)group,and from the stromal cells of patients with endometriosis,named the endometriosis(EMS)group.Nanopar-ticle tracking technology was used to detect the size,particle size distribution,and potential of exosomes in each group.West-ern blot was performed to analyze the expression of exosome markers CD9 and CD63 in the CON and EMS groups.The exosomes from the CON and EMS groups were co-cultured with THP-1 macrophages and named the CON-exosomes group and EMS-exosomes group,respectively.The PBS-Control group was used as a blank control and treated with an equal amount of PBS.MTT assay was used to evaluate the proliferation activity of the three groups of cells.Flow cytometry was performed to assess the apoptosis rate,as well as the ratio of M1-polarized(CD86+)and M2-polarized(CD206+)macrophages in each group.Fluorescent dye CM-Dil was used to label exosomes,DAPI was used to stain cell nuclei,and confocal microscopy was used to observe the phagocytic ability of macrophages towards exosomes.Results There was no significant difference in the physical parameters of exosomes between the CON and EMS groups(both P>0.05),with diameters ranging from 95.67 to 119.25 nm and 94.82 to 121.33 nm,and peak size of particle size distribution at 103.46 nm and 106.89 nm,respectively.The Zeta potentials of exosomes in the CON and EMS groups were(-29.28±3.91)mV and(-28.64±4.52)mV,respectively(P>0.05).There was no significant difference in the expression of exosome markers CD9 and CD63 between the two groups(P>0.05).Compared to the PBS-Control group,the THP-1 cell proliferation activity and M2 polarization ratio decreased in the CON-exosomes group(P<0.05),while the apoptosis rate showed no significant difference(P>0.05).Compared to the CON-exosomes group,the cell proliferation activity and M2 polarization ratio increased,and the apoptosis rate increased in the EMS-exosomes group(P<0.05).Compared to the CON-exosomes group,the THP-1 cell phagocytic function towards exosomes was weakened in the EMS-exosomes group.Conclusion Exosomes derived from endometriosis inhibit the prolif-eration activity and M2 polarization of THP-1 cells,promote apoptosis,and weaken their phagocytic function.
