Effect of hesperidin on liver injury in pregnant diabetes rats by regulating STAT3/SOCS3 signaling pathway
Objective To investigate the effect of hesperidin(HSP)on liver injury in gestational diabetes mellitus rats by regulating signal transducer and activator of transcription 3(STAT3)/suppressors of cytokine signaling 3(SOCS3)signaling pathway.Methods A GDM rat model was established.The rats were separated into a Control group,a Model group,low,me-dium,and high-dose hesperidin groups,and a high-dose hesperidin+STAT3 activator group.The pathological changes of liver tissue of rats in each group were observed.The levels of fasting blood glucose(FBG)and fasting insulin(FINS)were meas-ured,and insulin resistance(HOMA-IR)was calculated.Liver index,transaminase(ALT),aspartate transaminase(AST),malondialdehyde(MDA),and total superoxide dismutase(T-SOD)levels were detected.The expression of related proteins was detected.Results Compared with the Control group,the liver cells of rats in the Model group were arranged irregularly and disorderly,with inflammatory cell infiltration and necrosis,the levels of FBG,FINS,HOMA-IR,liver index,AST,ALT,and MDA were elevated,the expression of P-STAT3/STAT3 and SOCS3 were upregulated,and the level of T-SOD was de-creased(P<0.05).Compared with the Model group,the liver cell edema in the HSP-L,HSP-M,and HSP-H groups was im-proved,arranged neatly,and inflammatory cell infiltration was significantly reduced,the levels of FBG,FINS,HOMA-IR,liver index,AST,ALT,and MDA were decreased,the expression of P-STAT3/STAT3 and SOCS3 were downregulated,and the level of T-SOD was increased(P<0.05).Conclusion Hesperidin alleviates liver injury in GDM rats by inhibiting the STAT3/SOCS3 signaling pathway.
