橙皮素调节STAT3/SOCS3信号通路对妊娠糖尿病大鼠肝脏损伤的影响

目的 探究橙皮素(HSP)调节信号转导与转录激活子3(STAT3)/细胞因子信号转导抑制因子3(SOCS3)信号通路对妊娠糖尿病(GDM)大鼠肝脏损伤的影响.方法 建立GDM大鼠模型.将大鼠分为对照组、模型组、橙皮素低剂量处理(HSP-L)组、橙皮素中剂量处理(HSP-M)组、橙皮素高剂量处理(HSP-H)组和橙皮素高剂量+STAT3激活剂组.观察每组大鼠肝脏组织病理情况;检测空腹血糖(FBG)、空腹胰岛素(FINS)水平,计算胰岛素抵抗(HOMA-IR);检测肝脏指数、丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、丙二醛(MDA)、总超氧化物歧化酶(T-SOD)水平;检测相关蛋白表达.结果 与Control组相比,Model组大鼠肝细胞排列无规则、凌乱,有炎性细胞浸润和坏死,FBG、FINS、HOMA-IR、肝脏指数、AST、ALT、MDA 升高,p-STAT3/STAT3、SOCS3 上调表达,T-SOD水平降低(P<0.05);与Model组比较,HSP-L、HSP-M、HSP-H组肝细胞水肿有所改善,排列整齐,炎性细胞浸润明显减少,FBG、FINS、HOMA-IR、肝脏指数、AST、ALT、MDA 降低,p-STAT3/STAT3、SOCS3 下调表达,T-SOD 水平升高(P<0.05).结论 橙皮素抑制STAT3/SOCS3信号通路,可缓解GDM大鼠肝脏损伤.
Effect of hesperidin on liver injury in pregnant diabetes rats by regulating STAT3/SOCS3 signaling pathway
Objective To investigate the effect of hesperidin(HSP)on liver injury in gestational diabetes mellitus rats by regulating signal transducer and activator of transcription 3(STAT3)/suppressors of cytokine signaling 3(SOCS3)signaling pathway.Methods A GDM rat model was established.The rats were separated into a Control group,a Model group,low,me-dium,and high-dose hesperidin groups,and a high-dose hesperidin+STAT3 activator group.The pathological changes of liver tissue of rats in each group were observed.The levels of fasting blood glucose(FBG)and fasting insulin(FINS)were meas-ured,and insulin resistance(HOMA-IR)was calculated.Liver index,transaminase(ALT),aspartate transaminase(AST),malondialdehyde(MDA),and total superoxide dismutase(T-SOD)levels were detected.The expression of related proteins was detected.Results Compared with the Control group,the liver cells of rats in the Model group were arranged irregularly and disorderly,with inflammatory cell infiltration and necrosis,the levels of FBG,FINS,HOMA-IR,liver index,AST,ALT,and MDA were elevated,the expression of P-STAT3/STAT3 and SOCS3 were upregulated,and the level of T-SOD was de-creased(P<0.05).Compared with the Model group,the liver cell edema in the HSP-L,HSP-M,and HSP-H groups was im-proved,arranged neatly,and inflammatory cell infiltration was significantly reduced,the levels of FBG,FINS,HOMA-IR,liver index,AST,ALT,and MDA were decreased,the expression of P-STAT3/STAT3 and SOCS3 were downregulated,and the level of T-SOD was increased(P<0.05).Conclusion Hesperidin alleviates liver injury in GDM rats by inhibiting the STAT3/SOCS3 signaling pathway.
