南京地区草莓根腐病病原菌的分离及鉴定

目的:探究江苏省南京市溧水区草莓基地草莓(Fragaria×ananassa Duch.)根腐病的致病菌.方法:对分离的菌种通过形态学鉴定及返接实验确定致病菌株,对致病菌株开展基因内转录间隔区(ITS)、肌动蛋白基因(ACT)、3-磷酸甘油醛脱氢酶基因(GAPDH)、β-微管蛋白基因(TUB2)、查尔酮合酶基因(CHS)多基因联合测序鉴定.结果:从草莓基地采集草莓萎蔫发病植株,采用组织分离法获得20株菌株,返接试验确定了3株致病菌株:NJLS2、NJLS6、NJLS7.基于多基因联合测序鉴定,结合病原菌形态学特征鉴定引起溧水区草莓根腐病的是暹罗炭疽菌(Colletotrichum siamense).结论:本研究为分析草莓根腐病病原菌生物多样性提供理论依据,对南京溧水区草莓基地高效防治草莓根腐病具有实践意义.
Isolation and Identification of Pathogen of Strawberry Root Rot in Nanjing Area
Objective:Exploring the pathogenic bacteria of strawberry root rot in strawberry base,Lishui District,Nanjing City,Jiangsu Province.Methods:The isolated strains were determined by morphologi-cal identification and re-experiments.The pathogenic strains were identified by multigene sequencing with inner transcriptional spacer(ITS),actin genes(ACT),glyceraldehyde 3-phosphate dehydrogenase gene(GAPDH),β-tubulin gene(TUB2),and chalcone synthase gene(CHS).Results:Disease plants were collected from strawberry bases and 20 strains were obtained by tissue isolation.Three pathogenic strains NJLS2,NJLS6,and NJLS7 were identified by re-experiments.Based on the multigene combined sequencing and the morphological characteristics identification,Colletotrichum siamense was the patho-genic bacteria causing the strawberry root rot.Conclusion:This study provides a theoretical basis for ana-lyzing the biodiversity of strawberry root rot pathogens and has practical significance to the efficient pre-vention and control of strawberry root rot in Lishui strawberry base.
