Structural Characteristics and Phylogenetic Analysis of Chloroplast Genomes in Araceae Plants:Take 20 Species such as Arisaema heterophyllum for Example
Objective:Araceae plants were rich in resources and widely distributed.To study the chloro-plast genome structure of Araceae,so as to provide the basis for the phylogeny and evolution of Arace-ae.Methods:In this study,20 whole chloroplast genome sequences of this family were obtained from GenBank database as basic data,and the repetitive sequences,genomic differences,IR boundaries and phylogeny of these plants were analyzed by using REPuter,MISA,mVISTA and MAFFT software.Results:The results showed that the length of the chloroplast genome of Araceae plants ranged from 158 521 bp to 175 906 bp,encoding a total of 129 to 139 genes.In 20 Araceae species,50 repeat se-quences were detected,forward repeats and reverse repeats were the main types,and identified 253~482 SSR locis,mononucleotide and dinucleotide repeats were mostly.The global alignment of the se-quences showed that the differences in the non-coding regions were relatively large,and found three hypervariable regions accD,ycf2 and ycf1.Collinearity analysis showed that the genome sequence had high homology,rearrangements or inversions found in only one species.The IR boundary analysis showed that the Araceae plants were conserved to some extent.The screened SSR sites and hypervari-able regions can be used for genetic diversity analysis,species identification and DNA barcoding,etc.Conclusion:This study identifies the phylogenetic relationships of Araceae species,it provided a scien-tific basis for the study of species evolution and genetic diversity in Araceae.
