绉面草总黄酮提取工艺优化及美白活性研究

Optimization of Extraction Process and Whitening Activity of Total Flavonoids from Leucas zeylanica(L.)R.Br.

目的:优化绉面草总黄酮超声提取工艺,并考察其美白活性.方法:以绉面草总黄酮得率为指标,采用Box-Behnken响应面法对绉面草总黄酮提取工艺进行优化.通过绉面草总黄酮对DPPH、ABTS、·OH的清除能力以及总还原能力评价抗氧化活性,通过考察其对酪氨酸酶活性的抑制作用,并分析绉面草总黄酮对酪氨酸酶的抑制机理和抑制类型,评价其美白活性.结果:绉面草总黄酮最佳工艺条件为乙醇浓度65%,液料比18 mL/g,提取温度75℃,超声时间45 min,总黄酮得率高达107.36 mg/g;绉面草总黄酮对DPPH、ABTS以及·OH自由基具有良好的清除作用,并且总还原能力较好;同时,对单酚酶、二酚酶均有明显的抑制作用,抑制过程为不可逆的竞争与非竞争混合型抑制类型.结论:绉面草总黄酮提取工艺方法稳定可靠,且绉面草总黄酮具有良好的抗氧化能力和抑制酪氨酸酶活性,为绉面草在天然抗氧化剂以及祛斑美白类产品的开发利用提供了理论依据.
Objective:The ultrasonic extraction process of total flavonoids from Leucas zeylanica(L.)R.Br.was optimized and its whitening activity was investigated.Methods:The extraction process of to-tal flavonoids from L.zeylanica was optimized by Box-Behnken response surface method with the yield of total flavonoids as the index.The antioxidant activity of the total flavonoid extract was evaluat-ed by the scavenging capacity of DPPH,ABTS,·OH and the total reducing capacity.Evaluation of its whitening activity by investigating its inhibitory effect on tyrosinase activity,the inhibitory mechanism and inhibition type of total flavonoids of L.zeylanica were analyzed.Results:The optimal conditions for the total flavonoids of L.zeylantca were ethanol concentration of 65%,liquid-solid ratio of 18 mL/g,extraction temperature of 75℃,ultrasonic time of 45 min,and total flavonoids yield of 107.36 mg/g.The total flavonoids of L.zeylanica had a good scavenging effect on DPPH,ABTS and·OH free radi-cals,and the total reducing capacity was good.At the same time,the inhibition of monophenolase and diphenolase was obvious.The inhibition process was an irreversible mixture of competitive and non competitive inhibition types.Conclusion:Extraction process of total flavonoids from L.zeylanica was stable and reliable,and the total flavonoids of L.zeylanica had good antioxidant capacity and tyrosi-nase inhibitory activity,which provided a theoretical basis for the development and utilization of natu-ral antioxidants and whitening products.
