Objective:To clarify the effect of planting Rheum tanguticum on soil microbial community structure.Methods:High-throughput sequencing technology was used to analyze the characteristics of bacterial community structure in the soil at different depths(1-20 cm,21-40 cm and 41-60 cm)of R.tanguticum roots in different growth years(1-4 years).Results:A total of 2 080 bacterial opera-tional taxonomic units(OTUs)were obtained from the root soil of R.tanguticum in different growth years,belonging to 480 species,441 genera,263 families,172 orders,84 classes and 28 phyla.Pro-teobacteria,Acidobacteria,Actinobacteria,Chloroflexi and Gemmatimonadetes were the dominant groups with the relative abundance of the top 5 in the root soil of R.tanguticum,accounting for more than 0.80 of the entire bacterial community.The Mantel test results showed that soil available nitrogen,soil microbial biomass nitrogen(MBN),soil microbial biomass phosphorus(MBP)content and ni-trate reductase,cellulase and sucrase activities had extremely significant effects on bacterial communi-ty structure(P<0.01).Soil total phosphorus,available potassium content and catalase activity had sig-nificant effects on bacterial community structure(P<0.05).Affected by soil physical and chemical fac-tors,microbial biomass and enzyme activities,the number and diversity index of bacteria showed a'saddle type'trend of decreasing first and then increasing with the increase of the growth year of R.tanguticum,which was the lowest in the third year of growth.The results of NMDS analysis showed that the bacterial community structure in the second and third soil layers was significantly different from that of other samples in the third and fourth years of growth of R.tanguticum.In the FAPROTAX ecological function group,the abundance of chemoheterotrophic bacteria was the highest,reaching 24.0%-32.0%,followed by aerobic heterotrophic bacteria,accounting for 20.7%-28.2%,and the relative abundance of nitrifying bacteria was about 12.5%-18.1%.Conclusion:With the increase of the growth year of R.tanguticum,there was an indication that the succession of bacterial community structure and functional groups in soil occurred.
