白粉病对火棘叶片内生真菌群落结构的影响

Effects of Powdery Mildews on the Community Structure of Endophytic Fungi in Pyracantha fortuneana Leaves

目的:为比较健康和患白粉病火棘叶片内生真菌群落结构差异.方法:利用高通量测序技术对真菌r DNA-ITS(Internal transcribed space)序列进行测序分析.结果:两组样品共获得5门、16纲、3目、61科、83属、100种;健康样品HJF独有OTU为103个,感染白粉病的火棘叶片HJB独有OTU为19个,两个样品共有OTU为23个,HJF内生真菌物种丰度和多样性均高于HJB.物种分类显示,在门分类水平上,两组样品中优势菌群均为子囊菌门(Ascomycota);在科分类水平上,HJB中优势菌科为89.22%白粉菌科(Erysiphaceae);HJF中优势菌科为28.25%白粉菌科(Erysiphaceae)、12.30%丝孢酵母科(Trichosporonaceae)和12.31%Phaeosphaeriaceae;在属分类水平上,HJB中优势菌属为88.04%球针壳属(Phyllactinia);HJF中优势菌属为27.45%球针壳属(Phyllactinia)、12.03%Apiotri-chum、11.58%unclassified_f_Phaeosphaeriaceae.结论:健康组和罹病组火棘叶片的内生真菌群落结构及多样性存在明显差异,其优势、次优势菌属的相对丰度也存在明显差异,本研究明确了白粉病对火棘内生真菌结构的影响,为该病菌的致病机理和防控策略研究奠定基础.
Objective:In order to compare the endophytic fungal community structure between the healthy and the diseased leaves of P.fortuneana under natural conditions.Methods:ITS1 region of rD-NA-ITS genes of endophytic fungi were sequenced and analyzed using high-throughput sequencing technology.Results:The 5 phyla,16 classes,3 orders,61 families,83 genera,100 species were ob-tained from the two groups,and 103 OTUs unique to in healthy samples(HJF),19 OTUs unique in P.fortuneana leaves infected with powdery mildew(HJB),and 23 OTUs in two samples,the abundance and diversity of endophytic fungi species in HJF were higher than those in HJB.Species classification showed that Ascomycota was dominant in both of them at the phylum level.At the family level,89.22%Erysiphaceae were dominant in HJB.28.25%Erysiphacea,12.30%Trichosporonaceae and 12.31%Phaeosphaeriaceae were dominant in HJF.At the genus level,88.04%Phyllactinia were domi-nant in HJB.27.45%Phyllactinia,12.03%Apiotrichum and 11.58%unclassified_f_Phaeosphaeriace-aee were dominant in HJF.Conclusion:There were significant differences in the community structure and diversity of endophytic fungi between the healthy group and the infected group,as well as signifi-cant differences in the dominant and subdominant endophytic fungi relative abundance.
