超声辅助低共熔溶剂法优化提取珊瑚菌多糖及其抗氧化活性研究

摘要
目的:采用超声辅助低共熔溶剂法提取珊瑚菌中的多糖.方法:以Box-Behnken体系为基础,采用单因素结合响应面的方法进行四因素三水平试验设计优化珊瑚菌多糖的最佳提取工艺条件.结果:各因素对珊瑚菌多糖提取率的影响大小排序为:醇沉时间>液料比>提取温度>提取时间.以摩尔比1∶2的氯化胆碱—乙二醇体系作为提取剂,在提取温度60℃、液料比20∶1、超声时间40 min、醇沉时间24 h时的多糖提取率最高为31.76%.结论:采用超声辅助低共熔溶剂法提取珊瑚菌多糖比传统方法效果显著,可为珊瑚菌资源的开发利用提供理论参考.
Abstract
Objective:To extract polysaccharide from Ramaria botrytoides using ultrasound-assisted deep eutetic solvent method.Methods:Based on the Box-Behnken system,a four-factor three-level experimental design was carried out to optimize the extraction process conditions for R.botrytoides polysaccharide.Results:The influence of various factors on the extraction rate was as follows:alcohol precipitation time>liquid-to-material ratio>extraction temperature>extraction time.The highest extraction rate of 31.76% was achieved using a 1∶2 molar ratio choline chloride-ethylene glycol sys-tem as the extraction agent at an extraction temperature of 60°C,a liquid-material ratio of 20∶1,an ultrasonic time of 40 min,and an alcohol precipitation time of 24 h.Conclusion:The extraction of R.botrytoides polysaccharide by deep eutectic solvent method provided a reference for improving the ex-traction rate,which supplied a theoretical reference for making use of R.botrytoides.
