Objective:To extract polysaccharide from Ramaria botrytoides using ultrasound-assisted deep eutetic solvent method.Methods:Based on the Box-Behnken system,a four-factor three-level experimental design was carried out to optimize the extraction process conditions for R.botrytoides polysaccharide.Results:The influence of various factors on the extraction rate was as follows:alcohol precipitation time>liquid-to-material ratio>extraction temperature>extraction time.The highest extraction rate of 31.76% was achieved using a 1∶2 molar ratio choline chloride-ethylene glycol sys-tem as the extraction agent at an extraction temperature of 60°C,a liquid-material ratio of 20∶1,an ultrasonic time of 40 min,and an alcohol precipitation time of 24 h.Conclusion:The extraction of R.botrytoides polysaccharide by deep eutectic solvent method provided a reference for improving the ex-traction rate,which supplied a theoretical reference for making use of R.botrytoides.
