Objective:To explore the diversity of eukaryotic microalgae in some waters of Haixi Prefec-ture,Qinghai Province.Methods:8 samples from Chaerhan Salt Lake,Chaka Salt Lake,Gahai Salt Lake,Xiaochaidamu Salt Lake,Tuosu Lake,Keluke Lake,Dulan reservoir and Xueshanxiang were collected, and the community structure and diversity of eukaryotic microalgae were analyzed by high-throughput sequencing. Results:In the 8 samples,501 OTUs were detected, which belonged to 5 phy-la,23 classes,52 orders,89 families,139 genera, and contained 15 different OTUs.The 8 sampling points contained the most Chlorophyta, among which CKYH's dominant algae genus was Dunaliella.CEHYH's dominant algae genera were Mougeotia,Tetraselmis, and Dunaliella.DLSK and TSH's dominant algae genera were both Chlamydomonas and Tetraselmis.GHYH's dominant algae genera were Chlamydomonas and Biecheleria.KLKH's dominant algae genus was Tetraselmis.XCDMYH's dominant algae genera were Tetraselmis,Mantoniella, and Chlamydomonas. And XSX's dominant al-gae genera were Tetraselmis and Dunaliella. The difference analysis of the Alpha diversity indices showed that KLKH and DLSK had the highest richness, while CEHYH,XSX and DLSK had the high-est diversity.PCoA analysis showed that CKYH and CEHYH,TSH and XCDMYH were similar in community structure. The similarity of community structure between KLKH and XSX,as well as XSX and DLSK was small.GHYH was significantly different from the other 7 samples.Conclusion:Eukary-otic microalgae resources were abundant in Haixi Prefecture,Qinghai Province, even in extreme envi-ronments such as salt lakes and hot spring waters. The research results were of great significance for the future study on the adaptive evolutionary mechanism of algae species and the screening of functional al-gae species, and were conducive to the further development and utilization of eukaryotic microalgae re-sources.
