Brucea javanica was one of the traditional Chinese medicines, and its main active chemical com-ponents were quassionoids, which had pharmacological properties such as anti-tumor,anti-inflammatory,and antiviral. This article collected, summarized, and analyzed existing research results on quassionoids compounds,to explore the mechanisms and the structural differences of compounds with anti-inflammatory activity. The substitution of α,β-unsaturated ketones, glycosides, and olefinic bonds on the A,C,and D rings in quassionoids compounds could affect their anti-inflammatory activity. These quassionoids mainly exerted anti-inflammatory effects by regulating the intracellular NF-κB signaling pathway. In addition, they could also suppress the inflammatory response effectively by inhibiting the expression of various pro-inflammatory factors, including TNF-α,IL-6,IL-β,etc..The anti-inflammatory activity of quassionoids might be related to their anti-complement activity, particularly with complement proteins C3a and C5a.
