鸦胆子中苦木素类成分及其抗炎机制的研究进展

Research Advances of Anti-inflammatory Mechanism of Quassinoids in Brucea javanica

鸦胆子是我国传统中药之一,其主要活性化学成分是苦木素类化合物,具有抗肿瘤、抗炎、抗病毒等药理作用.本文通过整理、归纳和分析现有的苦木素类化合物研究成果,探讨具有抗炎活性的化合物结构的异同及其作用机制.苦木素类化合物中的A环、C环和D环上的α,β-不饱和酮、糖基、烯键的取代能影响其抗炎活性.这些化合物主要通过调控细胞内信号传导通路NF-κB发挥抗炎作用,此外,它们还能抑制多种促炎因子的表达,包括TNF-α、IL-6、IL-β等,从而有效地抑制炎症反应.苦木素类化合物的抗炎活性可能与其抗补体活性相关,特别是与补体蛋白C3a和C5a有关.
Brucea javanica was one of the traditional Chinese medicines,and its main active chemical com-ponents were quassionoids,which had pharmacological properties such as anti-tumor,anti-inflammatory,and antiviral.This article collected,summarized,and analyzed existing research results on quassionoids compounds,to explore the mechanisms and the structural differences of compounds with anti-inflammatory activity.The substitution of α,β-unsaturated ketones,glycosides,and olefinic bonds on the A,C,and D rings in quassionoids compounds could affect their anti-inflammatory activity.These quassionoids mainly exerted anti-inflammatory effects by regulating the intracellular NF-κB signaling pathway.In addition,they could also suppress the inflammatory response effectively by inhibiting the expression of various pro-inflammatory factors,including TNF-α,IL-6,IL-β,etc..The anti-inflammatory activity of quassionoids might be related to their anti-complement activity,particularly with complement proteins C3a and C5a.
