Objective:To study the potential application of extract from Kadsura coccinea leaf in the development of cosmetic raw materials.Methods:The extract of K.coccinea leaf was purified by mac-roporous adsorption resin,and the acute toxicity of the extract was studied by animal experiments.The anti-aging,whitening,and antiallergic effects of the extract were investigated by cell biological model experiments and chemical experiments.Results:AB-8 macroporous adsorption resin had a better puri-fication effect on the extract of K.coccinea leaf.The loading conditions were as follows:flavonoid con-centration of 1.0 mg/mL,pH 3,flow rate 3 BV/h,and loading volume 5 BV.The elution conditions were as follows:80%ethanol,with the same flow rate and elution volume as the loading condition.The tests of acute oral toxicity and acute dermal toxicity of the extract showed it was non-toxic.Within the range of 2.5-50.0 µg/mL,the extract could significantly raise the survival rates of skin fibroblasts damaged by H2 O2 and significantly reduce the levels of ROS and MDA,as well as the activities of SOD and CAT in damaged cells.Within the range of 0.1-2.0 mg/mL,the extract significantly reduced TYR activity in mouse B16 F10 melanoma cells.The extract had a significant inhibitory effect on HAase,with IC50 of 4.748 µg/mL.Conclusion:The extract of K.coccinea leaf purified by AB-8 macroporous adsorption resin had no acute toxicity,and showed anti-aging,whitening,anti-allergy and other ef-fects,which had certain development potential as a cosmetic raw material.
