Objective:Study on active sites and antibacterial activity of fenugreek were prepared by three-phase extraction.Methods:The three-phase extraction system was composed with different ratios of petroleum ether,acetonitrile,chloroform and water,and the active sites were prepared.The antibacterial activity was investigated by Oxford cup method,and the minimum inhibitory concentra-tion and minimum bactericidal concentration of Escherichia coli and Staphylococcus aureus were determined by two-fold dilution method.Results:Petroleum ether,acetonitrile,chloroform,water to system 1(5∶5∶1∶5)and system 2(5∶2∶5∶2)extract and ethanol extract 7 active sites.Compared with the blank control group,the F1 group inhibited S.aureus,The F2 to F5 group inbibited both E.coli and S.aureus.The lower layer of system 1 was the best antibacterial site,with good inhibi-tion effect on E.coli.The minimum inhibitory concentration and minimum bactericidal concentration for E.coli were 62.5 mg/mL and 125 mg/mL,respectively.And the minimum inhibitory concentration for S.aureus was 125 mg/mL,with no bactericidal effect.Conclusion:Three-phase extraction could effectively enrich the antibacterial active sites of fenugreek with obvious effect.
