蒙古族人群原发性高血压与AGT M235T及CYP11B2 C-344T基因多态性的相关性研究

摘要
目的 研究血管紧张素原(AGT)基因及醛固酮合成酶(CY11B2)基因多态性与蒙古族原发性高血压病的关系.方法 应用PCR-RFLP技术检测96例原发性高血压患者与正常对照组101名健康受试者AGT基因M235T多态性及CYP11B2基因C-344T多态性.结果 AGT的MM、MT、TT基因型频率在高血压组和正常血压组分别为0.21、0.73、0.06和0.26、0.65、0.09,差异无统计学意义(x2=0.495,P=0.482,OR=0.681,95%可信区间0.233~1.993);M、T等位基因的频率分别为0.58、0.42和0.59、0.41,差异无统计学意义(x2=0.051,P=0.821,OR=1.047,95%可信区间0.702~1.562);CYP11B2C-344T位点TT、TC、CC基因型的频率在高血压组和正常血压组分别为0.46、0.44、0.09和0.37、0.54、0.09,2组之间差异无统计学意义(x2=0.005,P=0.945,OR=1.034,95%可信区间0.392~2.728);T、C等位基因频率分别为0.69、0.31和0.64、0.36,差异无统计学意义(x2=0.928,P=0.335,OR=0.813,95%可信区间0.534~1.239);分析CYP11B2基因C-344T基因型与AGT基因M235T基因型时结果显示它们在蒙古族人群患高血压方面无协同作用.结论 AGT基因M235T位点与CYP11B2基因C-344T位点可能不是蒙古族人群患原发性高血压的遗传易感基因.
Abstract
Objective To investigate the association between gene ploymorphisms of angiotensinogen ( AGT) , aldosterone synthase ( CYP11B2) and essential hypertension in Mongolian patients. Methods M235T polymorphisms of AGT gene and C-344Tpolymorphisms of CYP11B2 gene were analyzed in 96 patients with essential hypertension and 101 healthy adults served as control using PCR-RFL P ( restriction fragment length polymorphism) . Results (1)MM, MT, TT genotypic frequency of AGT gene were 0.21, 0. 73 , 0. 06 in hypertension group and 0.26, 0. 65, 0.09 in healthy group. There was no significant difference in two groups(χ2 =0.495, P =0. 482,OR =0. 681,95% CI 0. 233 - 1. 993) . (2)the frequency of M,T allele were 0. 58, 0. 42 in hypertension group and 0. 59, 0. 41 in healthy group, there was no significant difference in two groups either(χ2 =0.051, P =0. 821 ,OR = 1. 047,95%CI 0. 702 - 1. 562). (3)TT,TC,CC genotypic frequency of CYP11B2 gene C-344 T site were 0. 46 、0.44 、0. 09 in hypertension group and 0. 37, 0. 54, 0. 09 in healthy group. There was no significant difference in two groups(χ2 =0.005, P =0. 945 ,OR = 1. 034,95% CI:0. 392 - 2. 728) . (4)the frequency of T,C allele were 0.69, 0. 31 in hypertension group and 0. 64, 0. 36 in healthy group. There was no significant difference in two groups( χ2 =0. 928, P =0. 335,OR = 0.813,95%CI:0. 534-1.239). (5)There was no synergistic action between C-344T genotype of CYP11 B2gene and M235T genotype of AGT gene in Mongolian people with essential hypertension . Conclusion M235T site of AGT gene and C-344T site of CYP11B2 gene were not inherent predisposing genes of Mongolian people with essential hypertension.
