UHPLC-MS/MS法测定小鼠血浆和脑中的4F-MDMB-BUTICA含量

目的:建立液相色谱-串联质谱法(UHPLC-MS/MS)测定小鼠血液和脑组织样本中4F-MDMB-BUTICA的含量.方法:采用液相色谱串联质谱法,使用C18 色谱柱,以0.1%甲酸水和乙腈为流动相,以5F-MDMB-PICA为内标物质,通过液-液萃取前处理后对滥用 4F-MDMB-BUTICA后小鼠的血液和脑组织样本中的 4F-MDMB-BUTICA进行定量检测,并对该方法进行方法学验证.结果:血浆和脑组织中的 4F-MDMB-BUTICA在 0.1~100 ng/ml的范围内均线性关系良好(R2 分别为 0.9942 和 0.9939),最低检出限为 0.1 ng/ml.日内准确度和日间准确度均在 85%~115%范围内,日内精密度和日间精密度均小于 15%,且基质效应的影响在 85%~115%范围内,表明该方法下基质无明显增强或抑制效应.结论:本研究建立了一种准确、灵敏、可靠的检测方法使用液相色谱串联质谱法(UHPLC-MS/MS)检测血液和脑组织样本中 4F-MDMB-BUTICA,为该物质基于动物的相关成瘾研究做基础.
Determination of 4F-MDMB-BUTICA in Mice Plasma and Brain by UHPLC-MS/MS
Objective:To establish a liquid chromatography tandem mass spectrometry(UHPLC-MS/MS)method for the determination of 4F-MDMB-BUTICA in mice blood and brain tissues.Methods:Liquid chromatography tandem mass spectrometry was used,with a C18 reverse phase chromatographic column and 0.1%formic acid water and acetonitrile as mobile phases.The liquid-liquid extraction was selected as pre-treatment method,and 5F-MDMB-PICA was used as the internal standard substance to quantitatively detect 4F-MDMB-BUTICA in blood and brain tissue samples of mice after an abuse of 4F-MDMB-BUTICA.The methodology was validated.Results:There was a good linear relationship between 4F-MDMB-BUTICA in plasma and brain tissue in the range of 0.1-100 ng/ml(R2 was 0.9942 and 0.9939,respectively),with a minimum detection limit of 0.1 ng/ml.The intra day accuracy and inter day accuracy were both between 85%and 115%,and intra day precision and inter day precision were less than 15%.The matrix effect was in the range from 85%to 115%,indicating that there was no significant enhancement or inhibition effect in this method.Conclusion:This study established an accurate,sensitive,and reliable detection method for 4F-MDMB-BUTICA in blood and brain tissue samples using liquid chromatography tandem mass spectrometry(UHPLC-MS/MS),laying the foundation for animal related addiction research on this substance.
