炮制对何首乌的药理与毒理作用的影响探究

目的:探究炮制对于何首乌药理和毒理作用的影响.方法:采用九蒸九晒法炮制何首乌,使用药典法炮制,采用高效液相色谱法在相应色谱条件下进样测定,分析何首乌2,3,5,4'-四羟基二苯乙烯葡萄糖苷(二苯乙烯苷)、大黄素、大黄素甲醚以及5-羟甲基糠醛(5-HMF)成分的含量,并以此分析药理成分和毒理成分的变化.结果:炮制前药理成分含量为(29.18±0.78)mg/g,炮制后含量为(29.52±0.98)mg/g,组间比较,差异无统计学意义(P>0.05).炮制前后大黄素比较,差异有统计学意义(P<0.05),而炮制前后大黄素甲醚比较,差异无统计学意义(P>0.05);炮制前生品中无5-HMF,经九蒸九晒法炮制后,其含量为0.0456 mg/g.结论:何首乌炮制品仍然具备良好的药理作用,并降低毒性成分含量,且5-HMF是区分生制品的关键,在临床应用上需要对两者严格区分,严格控制用药剂量,避免过度应用引发毒性反应.
Exploration on the Pharmacological and Toxicological Effects of Processing on Fleeceflower Root
Objective:To explore the effect of processing on the pharmacological and toxicological effects of fleeceflower root. Methods:Fleeceflower root was processed by Nine Steaming and Nine Sunning method,and processed by pharmacopoeia method. High performance liquid chromatography was used to determine the content of 2,3,5,4'-tetrahydroxystilbene glucoside (stilbene glucoside),emodin,emodin methyl ether and 5-hydroxymethylfurfural (5-HMF) in fleeceflower root under the corresponding chromatographic conditions,and the changes of pharmacological components and toxicological components were analyzed. Results:The contents of pharmacological components were (29.18±0.78) mg/g before processing and (29.52±0.98) mg/g after processing,and the differences were not statistically significant when comparing between groups (P>0.05). There was significant difference in the content of emodin before and after processing (P<0.05). There was no 5-HMF in the raw product before processing,and the content was 0.0456 mg/g after processing by Nine Steaming and Nine Sunning method.Conclusion:The processed products of fleeceflower root still have good pharmacological effects,reduce the content of toxic components. And 5-HMF is the key to distinguish raw products. In clinical application,it is necessary to strictly distinguish the two,strictly control the dosage,and avoid toxic reactions caused by excessive application.
