张氏除痹方调控miR-34a/SIRT1信号轴影响大鼠退变软骨细胞中炎症及细胞凋亡研究

目的:研究张氏除痹方对大鼠退变软骨细胞中炎症及细胞凋亡的影响及机制.方法:取大鼠关节软骨T25细胞,分别设置对照组、模型组、CBF组、CBF+miR-34a mimic组、CBF+miR-34a mimic+SIRT1组,模型组及CBF组使用IL-1β 建立退变软骨细胞模型,CBF+miR-34a mimic组转染miR-34a mimic并使用IL-1β 处理,CBF+miR-34a mimic+SIRT1组共转染过表达质粒SIRT1和miR-34a mimic,并使用IL-1β 处理.CBF组、CBF+miR-34a mimic组、CBF+miR-34a mimic+SIRT1组加入张氏除痹方含药血清.实时定量PCR测定T25细胞miR-34a水平,ELISA试剂盒测定ALP、BGP、SOD、MDA、GSH、IL-6、TNF-α水平,流式细胞术测定细胞凋亡率,免疫印迹法测定T25细胞SIRT1蛋白水平,双荧光素酶报告实验验证miR-34a与SIRT1靶向关系.结果:与模型组比较,CBF组细胞miR-34a、MDA、IL-6、TNF-α水平、软骨细胞凋亡率降低(P<0.05),软骨细胞SOD、GSH、ALP、BGP、SIRT1蛋白水平升高(P<0.05);双荧光素酶报告实验结果表明,与miR-34a NC组比较,miR-34a mimic组SIRT1 WT荧光素酶相对活性降低(P<0.05).结论:张氏除痹方能够显著抑制大鼠退变软骨细胞炎症反应,修复氧化应激损伤,抑制软骨细胞凋亡,其机制可能与调节miR-34a/SIRT1信号轴有关.
Study on the Effect of Zhang's Chubi Recipe on Inflammation and Apoptosis of Rat Degenerative Chondrocytes by Regulating miR-34a/SIRT1 Signaling Axis
Objective:To study the effect and mechanism of Zhang's Chubi Recipe on inflammation and apoptosis in degenerative chondrocytes of rats. Methods:Rat articular cartilage T25 cells were taken and divided into control group,model group,CBF group,CBF+miR-34 a mimic group,and CBF+miR-34 a mimic+SIRT1 group. IL-1β was used to establish a degenerative chondrocyte model in the model group and CBF group. The CBF+miR-34 a mimic group was transfected with miR-34 a mimic and treated with IL-1β. The CBF+miR-34 a mimic+SIRT1 group was co-transfected with overexpression plasmids SIRT1 and miR-34 a mimic. And treated with IL-1β. CBF group,CBF+miR-34 a mimic group and CBF+miR-34 a mimic+SIRT1 group were added with Zhang's Chubi recipe containing serum. The level of miR-34 a in T25 cells was determined by real-time quantitative PCR. The levels of ALP,BGP,SOD,MDA,GSH,IL-6 and TNF-α were determined by ELISA kit. The apoptosis rate was determined by flow cytometry. The level of SIRT1 protein in T25 cells was determined by Western blotting. The dual luciferase reporter assay was used to verify the targeting relationship between miR-34 a and SIRT1. Results:Compared with model group,the levels of miR-34a,MDA,IL-6,TNF-α,apoptosis rate of chondrocytes were decreased in CBF group (P<0.05),and the protein levels of SOD,GSH,ALP,BGP,SIRT1 were increased in chondrocytes (P<0.05). The results of double luciferase report showed that the relative activity of SIRT1 WT luciferase in miR-34a mimic group was decreased than that in miR-34a NC group (P<0.05). Conclusion:Zhang's Chubi recipe can significantly inhibit the inflammatory response of rat degenerative chondrocytes,repair oxidative stress injury,and inhibit chondrocyte apoptosis. The mechanism may be related to the regulation of miR-34a/SIRT1 signal axis.
