Experimental Study of Hawthorn Leaves Flavonoids on Peripheral Neuropathy of Diabetic Rats
Objective:To investigate the effect of hawthorn leaves flavonoids (HLF) on peripheral neuropathy (DPN) in diabetes rats. Methods:Diabetes rats induced by intraperitoneal injection of STZ were randomly divided into normal group,model group,low,medium and high dose HLF groups and mecobalamin group. After 12 weeks of administration,the thermal pain threshold and sciatic nerve conduction velocity of rats were measured,and changes in sciatic nerve and pancreatic tissue structure were observed. At the same time,body weight,fasting blood glucose,serum propylene glycol (MDA),and serum glutathione (GSH) were measured. Results:After 12 weeks of administration,the body weight,blood glucose,heat pain threshold,nerve conduction velocity,MDA and GSH levels in each group were significantly different from those in the normal group (P<0.05). The body weight of each HLF group was heavier than that of the model group (P<0.05),and the body weight of the HLF high and medium groups was heavier than that of the mecobalamin group (P<0.01). The blood glucose in each HLF group was lower than that in the model group and the mecobalamin group (P<0.01). The thermal pain threshold of HLF groups and mecobalamin group was lower than that of model group (P<0.01),and the thermal pain threshold of HLF low group was lower than that of mecobalamin group (P<0.01). MNCV and SNCV in HLF groups and mecobalamin group were higher than those in model group (P<0.05). MNCV in HLF low group was lower than that in mecobalamin group (P<0.01). SNCV in HLF medium and low groups was lower than that in mecobalamin group (P<0.05). MDA in HLF groups was lower than that in model group and mecobalamin group (P<0.05). GSH in HLF high and medium groups was higher than that in model group and mecobalamin group (P<0.01). Under the light microscope,the damage of sciatic nerve and pancreatic tissue structure in the model group and the mecobalamin group was obvious,and the damage in the HLF group was reduced. Conclusion:HLF can lower blood sugar,alleviate elevated pain threshold,reduce sciatic nerve conduction velocity,improve sciatic nerve and pancreatic tissue structure damage,and delay the occurrence and development of DPN.
