Expression and clinical significance of METTL7B in adult acute myeloid leukemia
Objective To investigate the differential expression and its clinical significance of METTL7B in adult acute myeloid leukemia(AML)based on the Cancer Genome Atlas(TCGA)and GEO data,and further predict its possible mechanism in AML.Methods The differential expression of METTL7B and survival progno-sis in adult AML tissues,as well as its association with drug response and immune cell infiltration were analyzed by integrating TCGA and GEO data from online bioinformatics databases.Functional enrichment analysis of MET-TL7B gene were performed.Results Compared with normal adult tissues,the expression of METTL7B in adult AML tissues was significantly increased(P<0.05).The high expression of METTL7B was significantly positively correlated with shorter overall survival and event-free survival in adult AML patients(P<0.05).The high expres-sion of METTL7B was significantly positively correlated with HDAC1 inhibitor resistance and the immune infiltra-tion of monocytes or macrophages(especially M2 macrophages)in AML(P<0.05).The METTL7B gene played a crucial role in regulating the biological processes of macrophage activation,and was involved in macrophage la-beling and regulating the interaction between immune cells and microRNAs in the tumor microen-vironment.Conclusions The expression of METTL7B is significantly up-regulated in adult AML tissues.Elevated expression of METTL7B is associated with poor prognosis in adult AML patients,suggesting its potential as a novel prognostic biomarker for AML.The METTL7B gene may participate in the progression of AML by mediating HDAC1 inhi-bitor resistance and M2 macrophage immune infiltration.
