Comparison of the effects of two administration routes of nimodipine on the Rho/ROCK signaling pathway in the basilar artery after subarachnoid hemorrhage in rabbits
Objective Nimodipine was administered to a New Zealand white rabbit with subarachnoid hemorrhage to investigate the effects of two different routes of administration on the posterior basilar artery Rho/ROCK signaling pathway.Methods A total of thirty-two New Zealand white rabbits were randomly divided into 4 groups (n=8):subarachnoid hemorrhage (SAH) group,intravenous nimodipine (NV) intervention group,Sham operation (Sham) group and nimodipine (ND) intervention group.Non anticoagulant autologous arterial blood (1 ml/kg) was injected into the ND group,SAH group,and NV group animals through the occipital cistern to replicate the SAH model.The sham group was injected with 0.9% sodium chloride injection (0.5 ml/kg) at 37℃through the oc-cipital cistern.Thirty minutes later,the SAH group was injected with 0.5 ml/kg of 2% nifedipine via the occipi-tal cistern,the NV group was injected with 0.5 ml/kg of 2% nifedipine via the rabbit ear vein,and the SAH and sham groups were injected with an equal amount of 0.9% sodium chloride injection (1 ml/kg).72 hours after SAH,neu-rological function scoring was performed,the expression of myosin light chain (MLC),Rho related kinase 2 (Rock2),and calmodulin (CAM) proteins in the basilar artery were detected by western blot.Results Compared with the sham group,the SAH group,ND group,and NV group showed upregulation of ROCK2 expression and decreased MLC/CAM expression.Compared with the SAH group,ROCK2 expression decreased and MLC/CAM ex-pression increased in ND group and NV group.Compared with the ND group,showed an in ROCK2 expression in-crease and MLC and CAM expression decreased in NV group.Conclusion Compared with intravenous injection,injection of nifedipine into the occipital cistern can more effectively affect the traditional Rho/ROCK signaling pathway in the basilar artery after SAH.
