目的:探讨GATA结合蛋白 3(GATA3)、性别决定区Y框蛋白 10 基因(SOX10)在乳腺癌不同分子分型中的表达特点及意义.方法:收集桂林市妇幼保健院病理科 2021 年 1 月—2022 年 12 月同时行SOX10 和GATA3 染色的乳腺癌标本 248 例,分析GATA3、SOX10 在乳腺癌中的表达与不同临床病理特征的关系.结果:GATA3 在乳腺癌管腔A(luminal A)型、管腔B(luminal B)型、人表皮生长因子受体 2(HER2)过表达型、三阴性乳腺癌(TNBC)中的阳性表达率分别为 100%、95.74%、93.75%、37.88%,型别间阳性表达率差异有统计学意义(P<0.05),SOX10 在乳腺癌luminal A型、luminal B型、HER2 过表达型、TNBC中的阳性表达率分别为 0、6.38%、9.38%、78.78%,型别间阳性表达率差异有统计学意义(P<0.05).联合应用SOX10、GATA3 在乳腺癌luminal A型、luminal B型、HER2 过表达型、TNBC中的阳性表达率分别为 100%、97.87%、96.88%、83.33%,虽型别间阳性表达率差异仍有统计学意义(P<0.05),但其在乳腺癌中阳性表达率高达 94.35%,明显高于单独GATA3 阳性表达率 81.05%和单独SOX10阳性表达率24.60%(P<0.05).GATA3+SOX10在原发性TNBC中阳性表达率高达84.48%(49/58),转移性TNBC中阳性表达率高达75.00%(6/8),两者比较差异无统计学意义(P>0.05).结论:GATA3 在乳腺癌luminal A型、luminal B型、HER2 过表达型中的阳性表达率较高,利于管腔型及HER2 过表达型乳腺癌病灶的评估和诊断,SOX10 在TNBC中的阳性表达率较高,有利于TNBC的诊断,联合应用SOX10及GATA3 可提高乳腺癌阳性检出率,且有利于转移性TNBC穿刺活检组织量少的样本诊断.
Expression and Significance of GATA3 and SOX10 in Different Molecular Types of Breast Cancer
Objective:To investigate the expression characteristics and significance of GATA binding protein 3(GATA3)and sex determination region Y-box protein 10(SOX10)in different molecular types of breast cancer.Method:A total of 248 breast cancer samples stained with both SOX10 and GATA3 were collected from Department of Pathology,Guilin Maternal and Child Health Hospital from January 2021 to December 2022.The relationship between the expression of GATA3 and SOX10 in breast cancer and different clinicopathological features was analyzed.Result:The positive expression rates of GATA3 in breast cancer luminal A,luminal B,HER2 overexpression and TNBC were 100%,95.74%,93.75%and 37.88%respectively,there was significant difference in the positive expression rate among types(P<0.05);the positive expression rates of SOX10 in breast cancer luminal A,luminal B,HER2 overexpression and TNBC were 0,6.38%,9.38%and 78.78%respectively,there was significant difference in the positive expression rate among types(P<0.05).The positive expression rates of SOX10 and GATA3 in breast cancer luminal A,luminal B,HER2 overexpression and TNBC were 100%,97.87%,96.88%and 83.33%respectively,although the positive expression rate among the types was still statistically significant(P<0.05),the positive expression rate in breast cancer was 94.35%,which was significantly higher than that of GATA3(81.05%)and SOX10(24.60%)(P<0.05).The positive expression rate of GATA3+SOX10 in primary TNBC was as high as 84.48%(49/58),and the positive expression rate in metastatic TNBC was as high as 75.00%(6/8),there was no statistical significance between them(P>0.05).Conclusion:GATA3 has a high positive expression rate in luminal A,luminal B,and HER2 overexpression types of breast cancer,which is conducive to the evaluation and diagnosis of luminal and HER2 overexpression types of breast cancer lesions,and SOX10 has a high positive expression rate in TNBC,which is conducive to the diagnosis of TNBC.The combined application of SOX10 and GATA3 can improve the positive detection rate of breast cancer,and is beneficial to the diagnosis of metastatic TNBC biopsy samples with small tissue volume.
