Effect of Blood Concentration of Olanzapine on the Levels of Prolactin,Glucose and Lipids in Patients with Schizophrenia
Objective:To observe the effects of blood concentration of Olanzapine on prolactin(PRL)and glucose and lipids levels in patients with schizophrenia.Method:A total of 86 patients with schizophrenia admitted to the Third People's Hospital of Yichun from December 2020 to December 2023 were selected as study subjects and divided into group A(daily dose<10 mg/d,n=23),group B(daily dose 10-19 mg/d,n=34)and group C(daily dose≥20 mg/d,n=29)according to different doses of Olanzapine.Serum levels of PRL,fasting blood glucose(FBG),total cholesterol(TC),triacylglycerol(TG),low density lipoprotein cholesterol(LDL-C)and high density lipoprotein cholesterol(HDL-C)at different time points were compared among three groups.Serum Olanzapine concentration was determined by high performance liquid chromatography before and after 4 and 8 weeks of treatment.Result:Before treatment,there were no significant differences in serum levels of PRL,FBG,TC,TG and LDL-C among three groups(P>0.05).After 4 and 8 weeks of treatment,serum levels of PRL,FBG,TC,TG,LDL-C and LDL-C in three groups were higher than those before treatment;after 8 weeks of treatment,serum levels of PRL,FBG,TC,TG and LDL-C in three groups were higher than those after 4 weeks of treatment,the differences were statistically significant(P<0.05).After 4 and 8 weeks of treatment,the levels of serum PRL,FBG,TC,TG and LDL-C in group A were lower than those in group B and group C,and those in group B were lower than those in group C,the differences were statistically significant(P<0.05).The total incidence of adverse reactions among the three groups was statistically significant(P<0.05).After 4 and 8 weeks of treatment,the blood concentration of Olanzapine in the three groups did not fluctuate significantly,and the differences were not statistically significant(P>0.05).After 4 and 8 weeks of treatment,the blood concentration of Olanzapine in the three groups was ranked as group A<group B<group C from low to high,and the differences were statistically significant(P<0.05).Pearson correlation coefficient analysis showed that Olanzapine serum concentration was significantly correlated with serum PRL,FBG,TC,TG and LDL-C(P<0.05).Conclusion:There is a significant correlation between Olanzapine serum concentration and serum PRL,glucose and lipid metabolism level in schizophrenia patients.The higher the serum concentration,the higher the serum PRL,glucose and lipid metabolism level.
