Objective To analyze the expressions of long non-coding RNA(lncRNA) ribonuclease P RNA component H1(RPPH1) and anterior gradient protein 3(AGR3) in colorectal cancer(CRC) tissues and their clinical significance. Methods A total of 98 patients with CRC who were admitted to XD Group Hospital from January 2017 to January 2020 were recruited.During the operation,CRC tissues and their paracancer tissues( at a distance of more than 3 cm from the edge of the cancer tissue, and no cancer cells being found by pathological examination) were collected. The expression levels of lncRNA RPPH1 were detected by using real-time quantitative polymerase chain reaction(PCR).The expression of AGR3 was detected by using immunohistochemical staining. The correlations of the expressions of lncRNA RPPH1 and AGR3 in the CRC tissues with the patients' clinicopathological features and survival prognosis were analyzed. Results The expression level of lncRNA RPPH1 in the CRC tissues was significantly higher than that in the paracancer tissues(P<0.05),and the positive rate of AGR3 in the CRC tissues was significantly higher than that in the paracancer tissues[65.31%(64/98) vs 10.20%(10/98),P<0.001].The expressions of lncRNA RPPH1 and AGR3 in the CRC tissues were significantly correlated with tumor TNM stage, tumor differentiation degree and lymph node metastasis(P<0.05).The survival prognosis of the low lcRNA RPPH1 expression group was significantly better than that of the high lcRNA RPPH1 expression group(log-rank test:x2=9.222,P=0.002).The survival prognosis of the AGR3-negative group was significantly better than that of the AGR3-positive group(log-rank test:x2=8.090,P=0.005).The results of multivariate Cox regression analysis showed that lymph node metastasis, tumor TNM stage Ⅲ,high expression of lncRNA RPPH1 and AGR3 positive were independent risk factors of poor survival prognosis for the CRC patients(P<0.05).Conclusion The increased expression level of lncRNA RPPH1 and positive expression of AGR3 in CRC tissues are associated with adverse clinicopathological features and poor prognosis of the patients.
