白驳丸及其拆方对白癜风小鼠CD8+T细胞及黑素细胞的影响

目的 探讨白驳丸及其拆方对白癜风小鼠CD8+T细胞及黑素细胞的影响.方法 正常C57BL/6小鼠作为空白组,同品系小鼠随机分为模型组、阳性药组、白驳丸及其拆方组.除空白组外,其余各组均构建内源性自身反应性CD8+T细胞靶向皮肤黑素细胞的白癜风动物模型.造模成功后,分别予以托法替尼、白驳丸及其拆方水溶液灌胃,空白组及模型组等量纯水灌胃,各组小鼠均灌胃10周.采用小型摄影棚及数码相机拍摄小鼠尾部皮肤,Image J图像处理后观察皮损脱色情况;采用全组织包埋免疫荧光观察皮损CD8+T细胞及黑素细胞数量及分布;采用流式细胞术观察皮损CD8+T细胞及黑素细胞比例及功能.结果 与空白组比较,模型组尾部皮损脱色面积增大,CD8+T细胞浸润及黑素细胞缺失增多,CD3+CD8+T、CD8+CD69+T、CD8+CXCR3+T细胞比例增加,CD45-CD117+黑素细胞比例下降.与模型组比较,白驳丸及其拆方可不同程度改善白癜风小鼠尾部皮损脱色情况,减少CD8+T细胞浸润及黑素细胞缺失,降低CD3+CD8+T、CD8+CD69+T、CD8+CXCR3+T细胞比例,增加CD45-CD117+黑素细胞比例;其中以白驳丸全方组效果最为显著,全方减养血活血通络药组效果最不明显.结论 白驳丸及其拆方可不同程度地抑制CD8+T细胞数量及功能,保护黑素细胞,改善白癜风小鼠尾部皮损脱色情况,其中养血活血通络药可能发挥核心作用.
Effects of Baibo Pills and its Modified Prescriptions on CD8+T Cells and Melanocytes in Vitiligo Mice
Objective To explore the effect of Baibo pills and its modified prescriptions on CD8+T cells and melanocytes in vitiligo mice.Methods Normal C57BL/6 mice were used as the blank group,and the mice of same strain were randomly divided into one model group,one positive drug group,and one Baibo pills and its modified prescription groups.Except for the blank group,all other groups constructed vitiligo animal models with autoreactive CD8+T cells targeting skin melanocytes.After successful modeling,tofacitinib,Baibo pills and its modified prescriptions were administered by gavage,while the blank group and model group were given equal amounts of pure water.All mice were administered by gavage for 10 weeks.Using a small studio and digital camera to capture the tail skin of mice,and observing the depigmentation of skin lesions after Image J image processing.Whole mount immunofluorescence was used to observe the number and distribution of CD8+T cells and melanocytes.Flow cytometry was used to observe the proportion and function of CD8+T cells and melanocytes.Results Compared with the blank group,the model group showed an increase in the depigmentation area of tail lesions,infiltration of CD8+T cells,and loss of melanocytes.The proportion of CD3+CD8+T,CD8+CD69+T,and CD8+CXCR3+T cells increased,while the proportion of CD45-CD117+melanocytes decreased.Compared with the model group,Baibo pills and its modified prescriptions could improve the depigmentation,reduce CD8+T cell infiltration and melanocyte deficiency,decrease the proportion of CD3+CD8+T,CD8+CD69+T,CD8+CXCR3+T cells,and increase the proportion of CD45-CD117+melanocytes to varying degrees.The effect of Baibo pills was the most significant,while the effect of Baibo Pills without nourishing blood,activating blood,and unblocking meridian herbs was the least significant.Conclusion Baibo pills and its modified prescriptions could inhibit the number and function of CD8+T cells,protect melanocytes,and reduce the depigmentation of tail skin lesions in vitiligo mice to varying degrees.Nourishing blood,activating blood,and unblocking meridian herbs may play a core role.
