摘要

目的 建立超高效液相色谱/四极杆飞行时间质谱与(NF-κB和β2-肾上腺素能受体)双荧光素酶报告基因检测系统相结合的方法,快速测定蛇胆陈皮口服液中抗炎平喘活性成分.方法 利用NF-κB和β2-肾上腺素能受体活性的双荧光素酶报告基因检测系统,筛选潜在的抗炎解痉成分,同时根据四级杆飞行时间质谱数据对筛选结构进行鉴定.结果 蛇胆陈皮口服液中辛弗林抑制β2-AR激活效应最明显;胆酸、甘氨胆酸、牛磺鹅去氧胆酸和牛磺胆酸抑制NF-κB激活作用最显著.结论 双活性评价谱效捕获方法是对筛选中药复方的有效成分的一种快速有力的手段.

Abstract

OBJECTIVE To develop a simple and dual-target method based on ultra-performance liquid chromatography/quadrupole time-of-flight mass spectrometry combined with dual-bioactive (NF-κB and β2-adrenergic receptor) luciferase reporter assay system for rapid determination of various bioactive compounds of the traditional Chinese medicine preparation Shedanchenpi oral liquid.METHODS Potential anti-inflammatory and spasmolytic constituents were screened using NF-κB and β2-adrenergic receptor activity luciferase reporter assay system and simultaneously identified according to the time-of-flight mass spectrometry data.RESULTS Synephrine had the most obvious inhibition on the activation effect of β2-AR.Cholalic acid,glycocholic acid,taurochenodeoxycholic acid,and taurocholic acid activation displayed the most significant inhibitory effect on the activation effect of NF-κB.CONCLUSION Dual bioactivity-integrated online spectrum efficiency capture technology is a powerful tool for the improved screening and identification of potential dual-target lead compounds in complex herbal medicines.
