强力枇杷露治疗慢性支气管炎模型小鼠作用机制研究

目的 探讨强力枇杷露治疗慢性支气管炎模型小鼠的作用机制.方法 将 72 只NIH小鼠随机分为空白对照组(A组,12 只,等体积纯净水)和模型组(60 只),采用烟熏联合脂多糖吸入法复制慢性支气管炎模型小鼠建模 45d后,将模型组小鼠随机分为模型对照组(B组,等体积纯净水),盐酸氨溴索组(C组,10 mL/kg),强力枇杷露低、中、高剂量组(D1 组、D2 组、D3 组,7.5,15,30 mL/kg),各12 只,雌雄各半.各组小鼠按 30 mL/kg灌胃给药,每日 1 次,连续 14 d.末次给药 24h后,采集小鼠血清,测定白细胞介素 6(IL-6)、白细胞介素 8(IL-8)水平;取右肺组织,测定肿瘤坏死因子-α(TNF-α)、丙二醛(MDA)及超氧化物歧化酶(SOD)含量;对左肺支气管组织进行病理组织学检查.结果 与B组比较,D1 组小鼠肺组织MDA含量显著降低(P<0.01);D2 组小鼠血清IL-8 的水平、肺组织MDA含量均显著降低(P<0.05);D3 组小鼠血清IL-8 水平、肺组织TNF-α和MDA含量均显著降低(P<0.05),SOD含量显著升高(P<0.05).与C组比较,D1 组小鼠血清IL-8 水平显著升高(P<0.05),肺组织SOD含量显著降低(P<0.05).病理组织学染色结果显示,与B组比较,D1 组、D2 组、D3 组小鼠慢性支气管炎病变程度减轻.结论 强力枇杷露治疗慢性支气管炎模型小鼠的作用机制可能与降低IL-8,TNF-α,MDA含量和升高SOD含量相关.
Mechanism of Qiangli Pipa Syrup in the Treatment of Chronic Bronchitis in Model Mice
Objective To investigate the mechanism of Qiangli Pipa Syrup in the treatment of chronic bronchitis in model mice.Methods A total of 72 NIH mice were randomly divided into the blank control group(group A,12 mice,equal volume of pure water)and the model group(60 mice).The mice in the model group were smoked and inhaled lipopolysaccharide to establish the chronic bronchitis mice models.After 45 d of modeling,the mice in the model group were randomly divided into the model control group(group B,equal volume of pure water),the ambroxol hydrochloride group(group C,10 mL/kg),and low-,medium-,and high-dose groups of Qiangli Pipa Syrup(groups D1,D2,D3,7.5,15,30 mL/kg),with 12 mice(half male and half female)in each group.Mice in each group were given gastric gavage at a dose of 30 mL/kg once daily for 14 d.At 24 h after the last administration,the serum of mice was collected to measure the levels of interleukin(IL)-6 and IL-8,the right lung tissue was collected to determine the contents of tumor necrosis factor-α(TNF-α),malondialdehyde(MDA),and superoxide dismutase(SOD),and the histopathology of the bronchial and left lung tissues was tested.Results Compared with those in group B,the MDA content in the lung tissue in group D1 was significantly lower(P<0.01);the serum IL-8 level and the MDA content in lung tissue in group D2 were significantly lower(P<0.05);the serum IL-8 level and the TNF-α and MDA contents in lung tissue in group D3 were significantly lower(P<0.05),while the SOD content in lung tissue in group D3 was significantly higher(P<0.05).Compared with those in group C,the serum IL-8 level in group D1 was significantly higher(P<0.05),while the SOD content in lung tissue in group D1 was significantly lower(P<0.05).The histopathological staining result showed a reduction in the degree of chronic bronchitis in groups D1,D2,and D3 compared with that in group B.Conclusion The mechanism of Qiangli Pipa Syrup in the treatment of chronic bronchitis in model mice may be related to the reduction of IL-8,TNF,MDA contents,and the increase of SOD content.
