Optimization of Extraction Process of Paeonol and Paeoniflorin from Paeonia Rockii by the Box-Behnken Response Surface Methodology
Objective To optimize the extraction process of paeonol and paeoniflorin in Paeonia rockii.Methods Based on the single-factor experiment,the Box-Behnken response surface methodology was used to optimize the optimal extraction process with the material-to-liquid ratio,extraction time,and ethanol volume fraction as influencing factors,and with the total weight comprehensive score of the extraction rates of paeonol and paeoniflorin as the evaluation indexes,and the validation test was conducted.Results The optimal extraction process was 28 times of liquid-solid ratio,150 min of extraction time,and 52%of ethanol volume fraction.Under the process conditions,the extraction rate of paeonol was 0.49%(RSD = 1.33%,n = 3),and the extraction rate of paeoniflorin was 5.38%(RSD = 1.07%,n = 3).Conclusion The extraction process was stable and feasible,which can be used for the extraction of paeonol and paeoniflorin from Paeonia rockii.
