Effects of Three Traditional Chinese Medicine Ingredients on the Oxidized Low-Density Lipoprotein-Induced Human Aortic Endothelial Cells Injury
Objective To investigate the effects of three traditional Chinese medicine(TCM)ingredients(curcumin,phillyrin and resveratrol)on the oxidized low-density lipoprotein(Ox-LDL)-induced human aortic endothelial cells(HAECs)injury.Methods HAECs were divided into the blank control group(equal volume of basic culture medium),the solvent control group[equal volume of basic culture medium containing dimethyl sulfoxide(DMSO)],the Ox-LDL group(20 µg/mL Ox-LDL),the curcumin groups ①②③④(20 µg/mL Ox-LDL + 2.5 µmol/L,5 µmol/L,10 µmol/L,20 µmol/L curcumin),the phillyrin groups ①②③④(20 µg/mL Ox-LDL + 10 µmol/L,20 µmol/L,40 µmol/L,80 µmol/L phillyrin),the resveratrol groups ①②③(20 µg/mL Ox-LDL + 20 µmol/L,40 µmol/L,80 µmol/L resveratrol).Cell activity was detected by the methyl thiazolyl tetrazolium(MTT)method,the cell proliferation rate was calculated,the expression levels of microtubule-associated protein 1 light-chain 3(LC3)and cyclooxygenase-2(COX-2)mRNA were detected by the real-time fluorescence quantitative polymerase chain reaction(qPCR).Results Compared with that in the Ox-LDL group,the cell proliferation rate in the phillyrin group ④,all the curcumin groups and resveratrol groups significantly decreased(P<0.01 or P<0.05).Compared with those in the Ox-LDL group,the expression levels of LC3 mRNA in the curcumin group ③,phillyrin groups ②③④ and all the resveratrol groups significantly increased,and the expression level of COX-2 mRNA significantly decreased(P<0.01 or P<0.05).Conclusion Curcumin,phillyrin and resveratrol can inhibit the proliferation of HAECs induced by Ox-LDL,stimulate the autophagy of injured cells,and inbibit the expression of inflammatory factor COX-2.
