Determination of Kaempferol in Domesticated Forsythiae Fructus by HPLC
Objective To establish a high-performance liquid chromatography(HPLC)method for content determination of kaempferol in domesticated Forsythiae Fructus.Methods The chromatographic column was Shim-pack VP-ODS column(250 mm×4.6 mm,4.6 μm),the mobile phase was methanol-0.4%phosphoric acid aqueous solution(50∶50,V/V),the flow rate was 1.0 mL/min,the detection wavelength was 365 nm,the column temperature was 30℃,and the injection volume was 10 μL.Results The linear range of kaempferol was 0.01-0.5 mg/mL.The RSDs of precision,stability and repeatability tests were all lower than 5%.The average recovery rates of kaempferol was 97.30%,with the RSD of 2.13%(n = 6).The content of kaempferol in domesticated Forsythiae Fructus from eight villages of Yunxi County,Shiyan City,Hubei Province was in the range of 0.037 7 to 1.088 7 mg/g.Conclusion This method is sensitive,repeatable and accurate,which can be used for the content determination of kaempferol in domesticated Forsythiae Fructus.
