Study on Anti-Hypoxia and Anti-Fatigue Effects of Compound Shenji Mixture on Mice
Objective To investigate the anti-hypoxia and anti-fatigue effects of Compound Shenji Mixture on mice.Methods The anti-hypoxia ability of mice was detected by the closed normobaric hypoxia test and sodium nitrite poisoning test,the survival time of mice was recorded,the superoxide dismutase(SOD),malondialdehyde(MDA)and glutathione peroxidase(GSH-Px)levels in the brain and myocardial tissue of mice were detected.The anti-fatigue ability of mice was detected by the weight-loaded swimming test,the exhaustive swimming time of mice was recorded,and the adenosine triphosphate(ATP),urea nitrogen(BUN),muscle glycogen(MG),hepatic glycogen glycogen(Gly),lactate(LA),lactate dehydrogenase(LDH)levels in serum were detected.In the above three tests,50 KM mice were selected and randomly divided into the blank control group(equal volume of physiological saline),the positive control group(0.3 g/kg of Hongjingtian Capsules),the Compound Shenji Mixture low-,medium-and high-dose groups(referred to as the low-,medium-and high-dose groups,1.0,2.0,3.0 g/kg).The mice in each group were given an equal volume of physiological saline or corresponding drug(10 mL/kg)by gavage for 14 d once a day.Results Compared with those in the blank control group,the survival time in the positive control group,low-,medium-and high-dose groups in the closed normobaric hypoxia test significantly prolonged(P<0.05);the SOD level in brain tissue in the high-dose group,the GSH-Px level in brain tissue in the low-,medium-and high-dose groups significantly increased,the MDA level in brain tissue in the positive control group,low-,medium-and high-dose groups significantly decreased;the SOD level in myocardial tissue in the high-dose group,the GSH-Px level in myocardial tissue in the positive control group,low-,medium-and high-dose groups significantly increased,and the MDA level in myocardial tissue in the low-and high-dose groups significantly decreased(P<0.05).Compared with those in the blank control group,the survival time in the positive control group,low-,medium-and high-dose groups in the sodium nitrite poisoning test significantly prolonged(P<0.05);the SOD level in brain tissue in the low-and medium-dose groups,the GSH-Px level in brain tissue in the medium-and high-dose groups significantly increased;the SOD level in myocardial tissue in the positive control group and medium-dose group,the GSH-Px level in myocardial tissue in the medium-dose group significantly increased,and the MDA level in myocardial tissue in the positive control group,low-,medium-and high-dose groups significantly decreased(P<0.05).Compared with those in the blank control group,the exhaustive swimming time in the low-,medium-and high-dose groups in weight-loaded swimming test significantly prolonged;the serum ATP level in the high-dose group,the LDH level in the low-,medium-and high-dose groups,and the Gly level in the medium-dose group significantly increased;the serum BUN and LA levels in the positive control group and low-,medium-and high-dose groups significantly decreased(P<0.05).Conclusion Compound Shenji Mixture can improve the hypoxia and fatigue in mice.
