目的 为酒大黄(唐古特大黄)配方颗粒的质量评价提供参考。方法 采用超高效液相色谱(UPLC)法,色谱柱为CORTECS UPLC T3柱(100 mm×2。1 mm,1。6 μm),流动相为甲醇-0。05%磷酸水溶液(梯度洗脱),流速为 0。25 mL/min,检测波长为 265 nm,柱温为35℃,进样量为 1 μL。采用中药色谱指纹图谱相似度评价系统(2012 版)建立 21 批样品的UPLC指纹图谱,并进行相似度评价。采用聚类分析(CA)法、主成分分析(PCA)法、正交偏最小二乘-判别分析(OPLS-DA)法评价样品质量,并筛查标志性成分。结果 共标定并指认出 18 个共有峰。21 批样品的指纹图谱相似度均大于 0。950,3 种分析方法均将样品分为 3 类,OPLS-DA法分析显示,异莲花掌苷、白藜芦醇-4′-O-β-D-(6′′-O-没食子酰)葡萄糖苷、儿茶素、大黄酚、没食子酸、番泻苷A、莲花掌苷和大黄酚-1-O-β-D-葡萄糖苷为制剂的标志性成分。结论 该方法简单可行,可为酒大黄(唐古特大黄)配方颗粒的质量评价提供参考。
UPLC Fingerprint and Chemical Pattern Recognition of Wine-Processed Rhei Radix et Rhizoma(Rheum Tanguticum)Formula Granules
Objective To provide a reference for the quality evaluation of Wine-Processed Rhei Radix et Rhizoma(Rheum tanguticum)Formula Granules.Methods Ultra-high-performance liquid chromatography(UPLC)was adopted.The chromatographic column was CORTECS UPLC T3 column(100 mm×2.1 mm,1.6 μm),the mobile phase was methanol-0.05%phosphoric acid aqueous solution(gradient elution),the flow rate was 0.25 mL/min,the detection wavelength was 265 nm,the column temperature was 35℃,and the injection volume was 1 μL.The Similarity Evaluation System for Chromatographic Fingerprint of Traditional Chinese Medicine(2012 Version)was used to establish UPLC fingerprints of 21 batches of samples,and the similarity evaluation was conducted.The cluster analysis(CA),principal component analysis(PCA)and orthogonal partial least squares-discriminant analysis(OPLS-DA)were used to evaluate the quality of samples and screen the characteristic components.Results A total of 18 common peaks were marked and identified.The similarity of the fingerprints of 21 batches of samples was greater than 0.950.The samples were divided into three categories by the above three analytical methods.The OPLS-DA showed that isolindleyin,resveratrol-4′-O-β-D-(6′′-O-galloy)glucoside,catechin,chrysophanol,gallic acid,sennoside A,lindleyin and chrysophanol-1-O-β-D-glucoside were the characteristic components of the preparation.Conclusion This method is simple and feasible,which can provide a reference for the quality evaluation of Wine-Processed Rhei Radix et Rhizoma(Rheum tanguticum)Formula Granules.
